摘  要:目的探究铅暴露对肥胖小鼠侧脑室下区(SVZ)区损伤状况.方法60只SPF级C57小鼠随机分为普通饲料组和高脂饲料组,用高脂饲料诱导肥胖小鼠模型.将普通饲料组分为对照组和铅暴露组,将肥胖小鼠分为肥胖组和肥胖+铅暴露组.对照组和肥胖组给予正常饮水;铅暴露组和肥胖+铅暴露组给予250 mg/L的醋酸铅饮用水,染毒8周.应用糖水偏好实验检测小鼠抑郁行为;应用ICP-MS检测小鼠全血和SVZ中铅含量;应用Western blot检测脑源性神经营养因子(BDNF)的蛋白表达;免疫荧光共定位法检测侧脑室下区新生神经元数量;应用ELISA法检测SVZ中白介素17(IL-17)含量;应用SPSS 22.0进行统计学分析.结果饲喂高脂饲料7周后,高脂饲料组小鼠的平均体重超过正常小鼠体重的20%,肥胖小鼠模型造模完成;染毒8周后,与对照组比较,铅暴露组、肥胖+铅暴露组小鼠血液和SVZ中的铅含量显著升高;肥胖组和铅暴露组小鼠的糖水偏好率分别为58.63%和63.63%,显著低于对照组的66.67%;与对照组比较,肥胖组和铅暴露组小鼠SVZ中新生神经元数量和BDNF蛋白表达下降;肥胖组和铅暴露组小鼠SVZ中IL-17含量分别为[28.89±5.16μg/g prot]和[35.47±6.77μg/g prot],显著高于对照组的[21.92±2.60μg/g prot],差异具有统计学意义.与铅暴露组或肥胖组比较,肥胖+铅暴露组糖水偏好率(41.67%)明显下降,SVZ中新生神经元数目显著降低,SVZ中IL-17含量[44.80±6.95μg/gprot]显著升高.结论铅暴露可能通过使肥胖小鼠SVZ区IL-17的表达增加和BDNF表达下降,SVZ神经细胞受损,进而导致出现抑郁行为.Objective To investigate the damage effect of lead exposure on subventricular zone(SVZ)area in obese mice.Methods A total of 60 C57 male mice were randomly divided into common diet group and high fat diet group,and obesity mouse model was established by high fat diet.Then the mice with regular diet were randomly divided into the obesity+lead exposure group were given 250mg/L lead acetate by free drinking water for 8 weeks.The depressive behavior of mice was detected by sucrose preference test.ICP-MS was used to detect the lead content in the whole blood and SVZ of mice.The protein expression of brain-derived neurotrophic factor(BDNF)was detected by Western blot;Immunofluorescence colocalization was used to detect the number of new neurons in SVZ.The content of IL-17 in SVZ was detected by ELISA.SPSS22.0 was used for statistical analysis.Results After feeding high-fat diet for 7 weeks,the average body weight of mice in high-fat diet group exceeded 20%of that of mice with regular diet,which indicated that and the obese mouse model was succeed;After 8 weeks of exposure,compared with the control group,the lead content in blood and SVZ of mice in lead exposure group or obesity+lead exposure group increased significantly;The sucrose preference of mice in the obesity group and lead exposure group were 58.63%and 63.63%,respectively,significantly lower than that of the control group(66.67%);The number of newborn neurons in SVZ and expression of BDNF in the obesity group and lead exposure group were lower than that of control group;The content of IL-17 in obese or lead exposed mice were[28.89±5.16μg/g prot]and[35.47±6.77μg/g prot],respectively,significantly higher than that of the control group[21.92±2.60μg/gprot];Compared with the lead exposed group and the obese group,the sucrose preference(41.67%),number of newborn neurons in SVZ and expression of BDNF decreased,the content of IL-17[44.80±6.95μg/g prot]increased in obese+lead group.Conclusion Lead exposure may cause depressive behavior by increasing the expressi
