摘 要:目的探究piRNA在正常妊娠和妊娠期糖尿病(GDM)孕妇子代脐血外泌体中的表达情况,并分析其与子代糖代谢紊乱的潜在关系。方法收集正常孕妇子代和GDM孕妇子代脐血各5例,提取外泌体总RNA,采用转录组测序检测两组样本中piRNA的表达情况,设定筛选条件筛选出差异表达的piRNA,预测其靶基因,并进行功能和信号通路分析。结果在脐血外泌体中鉴定到114个已知piRNA。与正常妊娠组相比,GDM组中存在显著差异表达的piRNA共有7个,共靶向4693个基因。生物信息学分析表明,这些piRNA靶基因主要富集于胰岛素信号通路、Hippo、TGF-β、mTOR、FoxO和Wnt信号通路。结论在正常孕妇与GDM孕妇子代脐血外泌体中piRNA存在差异表达,piRNA影响子代糖代谢的具体机制有待深入研究。Objective To investigate the expression of piRNA in cord blood exosomes from offspring of normal pregnant women and women with gestational diabetes mellitus,and to explore the potential relationship between piRNA and disorders of glucose metabolism in the offspring.Methods Cord blood was collected from the offspring of normal pregnant women and women with gestational diabetes mellitus in five cases respectively.Extraction of total RNA from cord blood exosomes using a kit.The expression of piRNAs in both groups was detected by transcriptome sequencing and to screen out differentially expressed piRNAs.Target mRNAs of differentially expressed piRNAs were predicated with related software,and GO and KEGG annotation of differentially expressed piRNAs was performed.Results A total of 114 known piRNAs were identified in cord blood exosomes.Seven piRNAs were significantly differentially expressed in the gestational diabetes group compared to the normal pregnancy group,which targeted 4693 genes.Bioinformatics analysis showed that these piRNA target genes were mainly enriched in insulin signaling pathway,Hippo signaling pathway,TGF-a signaling pathway,mTOR signaling pathway,FoxO signaling pathway,and Wnt signaling pathway.Conclusions There is a differential expression of piRNA in the cord blood exosomes from the offspring of normal pregnant women and gestational diabetic women,and the specific mechanism by which piRNA affects offspring glucose metabolism needs to be further investigated.
