摘  要:目的研究小核核糖核蛋白多肽A1(Small nuclear ribonucleoprotein A1,SNRPA1)在肠癌组织中的表达及对辛伐他汀抗肿瘤活性的影响。方法通过TCGA数据库、CPTAC及HPA数据库分析SNRPA1在肠癌中的的转录水平和蛋白表达水平,并通过组织芯片验证肿瘤组织与正常组织表达差异。利用NCI-60肿瘤细胞药物活性数据分析SNRPA1与辛伐他汀的相关性。通过qPCR筛选及WB验证SNRPA1高表达细胞株RKO及相对低表达细胞株HCT-116并采用CCK8法检测其对辛伐他汀抗肿瘤活性的影响。结果TCGA数据、CPTAC数据表明肠癌组织中SNRPA1的转录和表达水平显著高于癌旁组织(P<0.0001)。组织芯片免疫组化染色结果可见SNRPA1在肿瘤组织中表达升高。NCI-60数据表明SNRPA1的表达水平与辛伐他汀的抗肿瘤活性呈负相关性(R=-0.64,P<0.0001)。CCK8结果表明RKO细胞对辛伐他汀敏感性较低(IC 50=18.3μM),HCT-116对辛伐他汀敏感性相对较高(IC 50=9.91μM)。结论SNRPA1在肠癌组织中异常高表达,辛伐他汀的抗肿瘤活性与SNRPA1的表达呈负相关,降低SNRPA1的表达可增强辛伐他汀的抗肿瘤活性。Objective To investigate the expression of small nuclear ribonucleoprotein A1(SNRPA1)in colorectal cancer tissues and its effect on the anti-tumor activity of simvastatin.Methods The transcriptional level and protein expression of SNRPA1 in colorectal cancer tissues were analyzed by TCGA database,CPTAC database and HPA database,and the differential expression of SNRPA1 between tumor tissues and adjacent normal tissues were verified by tissue microarray.The correlation between SNRPA1 and simvastatin was analyzed using NCI-60 tumor cell drug activity data.The high expression SNRPA1 cell line RKO and relatively low expression SNRPA1 cell line HCT-116 were screened by qPCR and WB,and the effect of SNRPA1 on the anti-tumor activity of Simvastatin was detected by CCK8 assay.Results The TCGA data and CPTAC data showed that the transcription and expression levels of SNRPA1 in colon cancer tissues were significantly higher than those in para-cancer tissues(P<0.0001).Immunohistochemical staining of tissue array showed that SNRPA1 expression increased in tumor tissues.The NCI-60 data showed that the expression level of SNRPA1 in colon cancer cells was negatively correlated with the anti-tumor activity of Simvastatin(R=-0.64,P<0.0001).CCK8 assay demonstrated that RKO cell was less sensitive to simvastatin(IC 50=18.3),and HCT-116 was more sensitive to simvastatin(IC 50=9.91).Conclusions SNRPA1 is highly expressed in colorectal cancer tissues,and the anti-tumor activity of simvastatin is negatively correlated with the expression of SNRPA1.Decreasing the expression of SNRPA1 may enhance the anti-tumor activity of simvastatin.
