非小细胞肺癌患者组织及外周血中CABYR的表达及意义

摘　　要：目的检测非小细胞肺癌(NSCLC)患者肺癌组织和外周血中钙离子结合酪氨酸磷酸化调节蛋白(CABYR)的表达水平,探讨CABYR作为非小细胞肺癌患者肿瘤标志物的临床意义。方法收集NSCLC患者肿瘤组织及其对应癌旁组织、肺癌患者和正常人外周血,同时收集食道癌患者肿瘤组织及其对应癌旁组织作为其他肿瘤对照,用实时荧光定量PCR检测肺癌组织和外周血中CABYR-a/b mRNA的表达;用免疫组织化学法(IHC)检测肺癌组织及其对应癌旁组织中CABYR的表达水平。结果免疫组化证实NSCLC组织中有CABYR的表达;恶性与癌旁组织CABYR-a/b相对表达比值大于1的患者,在NSCLC患者肿瘤组织中CABYR-a/b mRNA的表达率(58.33%)明显高于食道癌组织(12.5%);与正常人外周血相比,NSCLC患者外周血中CABYR-a/b mRNA的表达水平略上调,但无统计学意义(P>0.05);与293T细胞株比较,CABYR-a/b在肺癌细胞株A549和H1650中的表达显著增高(P<0.01)。结论CABYR在NSCLC患者肿瘤组织和外周血中的表达均上调,对NSCLC的诊断和免疫治疗具有一定的临床价值,可能作为NSCLC的生物标志物,为非小细胞肺癌的免疫治疗提供一个新的靶点。Objective To investigate the expression level of calcium-binding tyrosine phosphorylation regulatory protein(CABYR)in lung cancer tissues and peripheral blood of patients with non-small cell lung cancer(NSCLC),and to investigate the clinical significance of CABYR as a tumor marker in patients with NSCLC.Methods Tumor tissues from NSCLC patients and their corresponding paracancerous tissues,peripheral blood from lung cancer patients and normal people were collected,while tumor tissues from esophageal cancer patients and their corresponding paracancerous tissues were collected as controls for other tumors,and the expression of CABYR-a/b mRNA was detected by real-time fluorescence quantitative PCR in lung cancer tissues and peripheral blood;and the expression level of CABYR was detected by immunohistochemical method(IHC)in lung cancer tissues and their corresponding paracancerous tissues.Results Immunohistochemistry confirmed the expression of CABYR in NSCLC tissues;the relative expression ratio of CABYR-a/b between malignant and paraneoplastic tissues was greater than 1.The expression rate of CABYR-a/b mRNA in tumor tissues of NSCLC patients(58.33%)was significantly higher than that of esophageal cancer tissues(12.5%);compared with the peripheral blood of normal people,the peripheral blood of NSCLC patients CABYR-a/b mRNA expression level was slightly up-regulated but not statistically significant(P>0.05);CABYR-a\b expression was significantly higher in lung cancer cell lines A549 and H1650 compared with 293T cell lines(P<0.01).Conclusion CABYRexpression was upregulated in both tumor tissues and peripheral blood of NSCLC patients,which is of clinical value for the diagnosis and immunotherapy of NSCLC and may serve as a biomarker of NSCLC and provide a new target for immunotherapy of NSCLC.
