摘　　要：目的观察结直肠癌(CRC)组织中调节性T细胞(Tregs)占比及叉头转录因子(Foxp3)、白细胞介素10(IL-10)、转化生长因子β_(1)(TGF-β_(1))表达情况,并分析Tregs占比与CRC临床病理参数的关系。方法选取手术切除的CRC组织及正常结肠组织各24例份,采用流式细胞术测算两组织中Tregs占比,实时定量PCR法检测两组织中Foxp3、IL-10、TGF-β_(1)mRNA相对表达量,Spearman相关分析法分析Tregs占比与CRC临床病理参数的关系。结果与正常结肠组织比较,CRC组织Tregs占比增加(P<0.05);与正常结肠组织比较,CRC组织Foxp3、IL-10、TGF-β_(1)mRNA相对表达量增加(P均<0.05);Tregs占比与CRC肿瘤分期相关(P<0.05)。结论CRC组织中Tregs占比增加,Foxp3、IL-10、TGF-β_(1)表达升高,Tregs占比与患者肿瘤分期有关,Tregs可能通过调节Foxp3、IL-10、TGF-β_(1)发挥免疫逃逸作用。Objective To explore the proportion of regulatory T cells(Tregs)in colorectal cancer(CRC)tissues and the expression of Foxp3,interleukin 10(IL-10),and transforming growth factorβ_(1)(TGF-β_(1)),and to analyze the relationships between the proportion of Tregs and the clinicopathological parameters of CRC.Methods Twenty-four CRC tissues and 24 normal colon tissues were selected from surgical resection.Flow cytometry was used to detect the proportion of Tregs,and real-time quantitative PCR was used to detect the expression levels of Foxp3,IL-10 and TGF-β_(1)mRNA in these two groups.Spearman correlation analysis was used to analyze the relationships between the proportion of Tregs and the clinicopathological parameters of CRC.Results Compared with the normal colon tissues,the proportion of Tregs in CRC tissues increased(P<0.05).Compared with the normal colon tissues,the relative expression of Foxp3,IL-10 and TGF-β_(1)mRNA in CRC tissue increased(all P<0.05).Tregs proportion was correlated with the stage of CRC(P<0.05).Conclusions The proportion of Tregs in CRC tissues increased,and the expression of Foxp3,IL-10 and TGF-β_(1)increased.The proportion of Tregs was related to the tumor stage of patients,and Tregs played an immune escape role probably by regulating Foxp3,IL-10 and TGF-β_(1).
