高血压脑出血患者脑组织microRNA表达谱及生物信息学分析

摘  要:目的构建高血压脑出血患者脑组织差异表达miRNA,并进行生物信息学分析。方法选取行高血压脑出血手术患者脑血肿腔壁脑组织标本16例,同期选择重型颅脑损伤且具有高血压病史患者脑组织标本16例作为对照组。通过Trizol法提取标本总RNA,筛选出差异表达的miRNA。利用生物信息学技术分析差异表达miRNA的特点,通过String数据库得到靶基因蛋白互作分析结果,Cytoscape软件进行miRNA-mRNA核心调控网络可视化。结果实验组与对照组相比较共筛选出差异表达miRNA 43个(P<0.05),其中上调miRNA共7个,下调miRNA共36个。对差异miRNA的靶基因进行KEGG/GO富集分析,得到与高血压脑出血相关的信号通路:cAMP信号通路、ErbB信号通路、Calcium信号通路、Wnt信号通路(P<0.05)。构建miRNA-mRNA可视化网络图,发现关联度最高的miRNA为:hsa-miR-1303、hsa-miR-1972、hsa-miR-362-3p、hsa-miR-101-3p、hsa-miR-378d,PPI蛋白互作分析发现连接度最高的前三个基因为:MAPK1、GRB2和RAC1。结论差异miRNA可能在自发性高血压脑出血的发病机制中具有重要作用,可能为自发性高血压脑出血的治疗及预防提供潜在治疗靶点及方向。Objective To construct microRNA differentially expressed in hypertensive intracerebral hemorrhage patients and analyze it bioinformatically.Methods 16 patients with hypertensive intracerebral hemorrhage were selected,and 16 patients with severe craniocerebral injury and hypertensive history were selected as control group.Total RNA was extracted by Trizol method and differential expression miRNAs were screened.Cytoscape software was used to visualize the miRNA-mRNA core regulatory network.The characteristics of differentially expressed mirnas were analyzed by bioinformatics technology.The results of target gene protein interaction analysis were obtained by String database.Results Total of 43 differentially expressed miRNAs were detected between experimental group and control group(P<0.05),including 7 up-regulated miRNAs and 36 down-regulated miRNAs.KEGG/GO enrichment analysis was performed on the target genes of differential miRNA,and the signaling pathways associated with hypertensive intracerebral hemorrhage were obtained:cAMP signaling pathway,ErbB signaling pathway,Calcium signaling pathway,Wnt signaling pathway(P<0.05).The visualization network diagram of miRNA-mRNA was constructed,and it was found that miRNAs with the highest correlation were hsa-miR-1303,hsa-miR-1972,hsa-miR-362-3p,hsa-miR-101-3p and hsa-miR-378d.PPI showed that top 3 genes with the highest connectivity were MAPK1,GRB2 and RAC1.Conclusion Differential miRNA may play an important role in the pathogenesis of spontaneous hypertensive intracerebral hemorrhage,providing therapeutic targets and directions for the treatment and prevention of spontaneous hypertensive intracerebral hemorrhage.
