摘 要:目的探讨不同压强持续性CO_(2)气腹对结肠癌细胞体内侵袭能力的影响。方法选用人结肠癌细胞株SW1116,将细胞放置在含有10%小牛血清的RPMI1640培养液中,置入37℃、5%CO_(2)培养箱中常规培养,建立体外气腹模型,将SW1116细胞置入其中。将SW1116细胞分为5组,分别放置在不同CO_(2)气体压强环境中,即常规培养组(对照组:5%CO_(2)、37℃)、A组(6 mmHgCO_(2)气腹)、B组(9 mmHgCO_(2)气腹)、C组(12 mmHgCO_(2)气腹)、D组(15 mmHgCO_(2)气腹)各20份,维持1 h后取出细胞。在气腹模型试验结束后,继续常规培养24、48、72 h后对细胞进行体外细胞侵袭试验。结果对照组在经过气腹处理后,从气腹结束到72 h后,细胞侵袭荧光强度增强,差异均有统计学意义(P<0.05);A组随着时间的推移,荧光强度略微增加,B、C、D组随着时间的推移,变化并不显著;在经过气腹处理后即刻以及24、48、72 h后,D组结肠癌细胞侵袭荧光强度减弱,明显低于对照组、A组、B组、C组,差异均有统计学意义(P<0.05)。结论不同压强持续CO_(2)气腹对结肠癌细胞体内侵袭能力产生一定的影响,高压强CO_(2)气腹的建立对结肠癌细胞的侵袭能力产生了一定的抑制效果;CO_(2)气腹72 h内不同作用时间对结肠癌细胞的侵袭能力未见明显影响。Objective To explore the effect of sustained CO_(2)pneumoperitoneum with different pressure levels on the invasion ability of colon cancer cells in vivo.Methods The human colon cancer cell line SW1116 was selected and placed in RPMI1640 culture medium containing 10%calf serum.The cells were then placed in a 37℃,5%CO_(2)incubator for routine cultivation.An in vitro pneumoperitoneum model was established,and SW1116 cells were placed in it.SW1116 cells were divided into 5 groups and placed in different CO_(2)gas pressure environments,namely the conventional culture group(control group:5%CO_(2),37℃),group A(6 mmHgCO_(2)pneumoperitoneum),and group B(9 mmHgCO_(2)pneumoperitoneum).In group C(12 mmHgCO_(2)pneumoperitoneum),group D(15 mmHgCO_(2)pneumoperitoneum),the cells were taken out after maintaining for 1 hour.After the pneumoperitoneum model test,the cells were cultured routinely for 24,48,and 72 h,and then the cells were subjected to in vitro cell invasion tests.Results After pneumoperitoneum treatment,from the end of the pneumoperitoneum to 72 h,the fluorescence intensity of cell invasion in the control group increased,the difference was statistically significant(P<0.05).The fluorescence intensity of group A increased slightly over time,the changes in groups B,C and D were not significant over time.Immediately after the pneumoperitoneum treatment and 24,48,72 h,the colon cancer cell invasion fluorescence intensity of group D decreased,which was significantly lower than that of control group,group A,group B,group C,the difference was statistically significant(P<0.05).Conclusion Continuous CO_(2)pneumoperitoneum with different pressures has a certain impact on the invasion and metastasis ability of colon cancer cells in vivo,and the establishment of high-pressure and strong CO_(2)pneumoperitoneum has a certain inhibitory effect on the invasion ability of colon cancer cells.CO_(2)pneumoperitoneum has no significant effect on the invasive ability of colon cancer cells at different action times within 72 hours.
