摘 要:目前海参中的岩藻聚糖与硫酸软骨素的简便分离方法较少,该研究拟建立一种双水相分离方法用于海参中的岩藻聚糖与硫酸软骨素的简便高效分离。以海地瓜岩藻聚糖和海地瓜硫酸软骨素为研究材料,以分配系数、回收率为评价指标,进行单因素试验,分别考察盐种类、聚乙二醇(polyethylene glycol,PEG)分子质量、pH、PEG质量分数和K_(2)HPO_(4)质量分数对海地瓜岩藻聚糖及海地瓜硫酸软骨素在双水相体系中分配行为的影响。结果表明,PEG 6000-K_(2)HPO_(4)双水相体系可以分离海地瓜中的岩藻聚糖与硫酸软骨素,其中岩藻聚糖集中分配于上相,硫酸软骨素集中分配于下相。最佳分离条件为PEG 6000的质量分数为15%,K_(2)HPO_(4)的质量分数为10%。此条件下,海地瓜中的岩藻聚糖在该体系的分配系数为6.36,在上相的回收率为(86.92±0.97)%;硫酸软骨素在该体系的分配系数为16.53,在下相的回收率为(106.47±1.84)%。该研究建立的方法分离效果良好且简便快捷,具有良好的应用前景。There is a lack of simple and efficient separation method of sulfated fucan and chondroitin sulfate in sea cucumber.This study intends to establish a method based on polyethylene glycol(PEG)-salt aqueous two-phase system for the highly selective separation of sulfated fucan and chondroitin sulfate.Using the distribution coefficient and recovery rate as parameters,the effect of different salt types,PEG molecular weight,pH,concentration of PEG and K_(2)HPO_(4) on the partition behavior of sulfated fucan and chondroitin sulfate from Acaudina molpadioides was discussed.The sulfated fucan and chondroitin sulfate could be separated by the PEG-K_(2)HPO_(4) aqueous two-phase system.Sulfated fucan was majorly presented in the upper phase,while chondroitin sulfate distributed in the lower phase.Under the most favorable partition conditions with 15%PEG 6000 and 10%K_(2)HPO_(4),the partition coefficient and recovery rate of sulfated fucan were achieved 6.36 and(86.92±0.97)%.Meanwhile,the partition coefficient and recovery rate of chondroitin sulfate were achieved 16.53 and(106.47±1.84)%,respectively.The aqueous two-phase separation method established in this study could implement the effective separation of sulfated fucan and chondroitin sulfate from Acaudina molpadioides.This method can be applied to the separation of sulfated fucan and chondroitin sulfate of other sea cucumbers species,which has a good application prospect.
