摘 要:为探究增强Debaryomyces nepalensis控制冬枣采后病害的方法,采用果实及体外实验研究了CaCl_(2)和D.nepalensis复合处理对冬枣采后黑斑病的控制效果和机制。结果表明,所选质量浓度为5、10、20、40 g/L的CaCl_(2)对链格孢菌(Alternaria alternata)没有直接抑菌作用;5 g/L CaCl_(2)结合1×10^(8)CFU/mL的D.nepalensis处理后,能够控制冬枣采后黑斑病,且控制效果显著强于D.nepalensis单独处理。5 g/L CaCl_(2)结合1×10^(8)CFU/mL的D.nepalensis处理,能够显著增强D.nepalensis在冬枣果实伤口处的定殖能力,促进酵母生长并增强D.nepalensis生物膜形成能力。除此之外,CaCl_(2)结合D.nepalensis处理后,冬枣果实苯丙氨酸解氨酶、肉桂酸-4-羟基化酶、4-香豆酸辅酶A连接酶和过氧化物酶活性增加,同时,总酚、类黄酮及木质素含量显著上升。综上,CaCl_(2)通过增强D.nepalensis定殖能力及果实抗性提高D.nepalensis对冬枣采后黑斑病的控病能力。In order to investigate the enhancement of Debaryomyces nepalensis for postharvest disease control of jujube,the effect and mechanism of combined treatment of CaCl_(2)and D.nepalensis on postharvest black spot disease control of jujube were studied by fruit and in vitro experiments.The results showed that the selected concentrations(5,10,20,40 g/L)of CaCl_(2)had no direct inhibitory effect against Alternaria alternata.However,the treatment of 5 g/L CaCl_(2)combined with 1×10^(8)CFU/mL of D.nepalensis was able to control postharvest black spot disease of jujube,and the control effect was significantly better than that of D.nepalensis alone.The treatment of 5 g/L CaCl_(2)combined with 1×10^(8)CFU/mL of D.nepalensis significantly enhanced the ability of D.nepalensis to colonize on the wound of jujube,promoted yeast growth and enhanced the biofilm formation of D.nepalensis.In addition,CaCl_(2)in combination with D.nepalensis treatment significantly enhanced the growth of yeast and biofilm formation of D.nepalensis.The activity of phenylalanine deaminase(PAL),cinnamate-4-hydroxylase(C4H),4-coumaric acid coenzyme-A-ligase(4CL)and peroxidase(POD)increased,and total phenol,flavonoid and lignin contents increased significantly.In conclusion,CaCl_(2)improved the disease control capacity of D.nepalensis against postharvest black spot disease of jujube by enhancing the colonization ability and fruit resistance of D.nepalensis.
