摘 要:本试验比较了AB-8、NKA-9、HPD-100、DM130、D101及X-5六种大孔树脂对青蒿总多酚的纯化效果,筛选出富集青蒿多酚的最佳大孔树脂型号为HPD-100。采用单因素试验确定其最佳工艺参数:上样液pH值为2.0,质量浓度为50 mg/mL,上样量为13 BV,静态吸附2 h;洗脱液是体积分数为70%乙醇溶液,用量为5 BV。纯化后的青蒿总多酚纯度较纯化前增加了1.63倍。纯化后的青蒿总多酚对RSV、HSV-1和HSV-2均表现出显著的体外抗病毒活性,而对EV71未表现出较好的抑制效果。In this experiment,the purification effects of AB-8,NKA-9,HPD-100,DM130,D101 and X-5 macroporous resins on the polyphenol of Artemisia annua Linn.were compared.The optimal macroporous resin for enrichment of A.annua.polyphenol was selected as HPD-100.The optimal process parameters were determined by single factor tests:pH value of loading solution as 2.0,mass concentration as 50 mg/mL,loading volume as 13 BV,static adsorption for 2 h,and 5 BV of 70%ethanol solution as eluent.After purification,the purity of A.annua polyphenol increased by 1.63 times compared with that before purification.The purified A.annua polyphenol showed significant antiviral activities against RSV,HSV-1 and HSV-2 in vitro,but did not show good inhibitory effect against EV71.
