姜黄素抑制脊髓组蛋白甲基化在大鼠急性疼痛中的作用

摘  要:目的:探讨姜黄素对大鼠急性疼痛及脊髓组蛋白甲基化表达的作用。方法:采用随机数字表法将成功鞘内置管的48只健康雄性6~8 w Wistar大鼠(200~250 g)分成四组(n=12):对照组(D组),疼痛模型组(I组),姜黄素组(C组),二甲基亚砜组(DMSO组)。I组、C组、DMSO组采用左后足切口法制备急性疼痛模型。制备疼痛模型前30 min,D组、I组鞘内注射生理盐水10μL,C组注射姜黄素10μL(50μg·μL^(-1)),DMSO组注射二甲基亚砜10μL。分别于术前24 h(T_(0))、术后1 h(T_(1))、3 h(T_(2))、6 h(T_(3))测量大鼠左后足的机械性痛阈(Mechanical paw withdrawal threshold,MWT)及热痛阈(Thermal paw withdrawal latency,TWL);分别于T_(1)、T_(3)痛阈测定取脊髓组织,采用免疫组化测定组蛋白甲基化表达水平。结果:与D组比较,I组、C组、DMSO组T_(1-3)时MWT下降、TWL缩短,T_(1)、T_(3)时组蛋白甲基化表达上调(P<0.05)。与I组比较,C组T_(1-3)时MWT上升、TWL延长,T_(1)、T_(3)时组蛋白甲基化表达上调(P<0.05);DMSO组无统计学差异(P>0.05)。结论:姜黄素可以减轻大鼠的急性疼痛,并降低了脊髓组蛋白甲基化水平的表达。Objective:To investigate the effect of Curcumin on acute pain and expression of Histone methylation in the spinal cord of rats.Methods:Forty-eight healthy male 6-8 w Wistar rats(200-250 g)with successful intrathecal catheterization were randomly divided into four groups(n=12):the control group(group D),pain model group(group I),Curcumin group(group C),and Dimethyl sulfoxide group(DMSO group).The rats in Group I,C,and DMSO were used to establish acute pain models using the left posterior foot incision method.Thirty minutes before preparing the pain model,the rats in groups D and I were injected with physiological saline intrathecally for 10μL.Group C was injected curcumin 10μL(50μg·μL^(-1)).In the DMSO group,10μL DMSO was injected.The mechanical paw with drawn threshold(MWT)and thermal paw with drawn latency(TWL)of the left hind foot of rats were measured 24 hours before surgery(T_(0)),1 hour after surgery(T_(1)),3 hours after surgery(T_(2)),and 6 hours after surgery(T_(3)).Spinal cord tissue was taken at T_(1)and T_(3)pain threshold respectively,and the expression level of histone methylation was measured by immunohistochemistry.Results:Compared with group D,MWT decreased and TWL shortened at T_(1-3)in group I,group C and DMSO,and the expression of Histone methylation was up-regulated at T_(1)and T_(3)(P<0.05).Compared with group I,MWT increased and TWL prolonged in group C at T_(1-3),and the expression of Histone methylation was up-regulated at T_(1)and T_(3)(P<0.05).There was no statistically significant difference in the DMSO group(P>0.05).Conclusion:Curcumin can alleviate acute pain in rats and reduce the expression of Histone methylation in the spinal cord.
