摘 要:利用高效液相色谱法对油莎豆块茎(茎豆)萌发过程中(干茎豆-吸胀茎豆-初发芽茎豆)多种内源激素的含量进行检测,旨在揭示内源激素含量的变化规律及调控作用。结果表明,脱落酸(ABA)、生长素(IAA)和茉莉酸(JA)的含量在油莎豆休眠的干茎豆中较高,随后均呈先下降后上升的趋势;相反,赤霉素(GA 3)和玉米素核苷(ZR)的含量起始较低,随后均呈先上升后下降的趋势,表明ABA、IAA和JA为萌发抑制类激素而GA 3和ZR为萌发促进类激素,并通过相应的外源激素处理进行了验证。在此基础上进一步研究各种激素之间的协同途径,将为解析油莎豆茎豆休眠与萌发的调控机制奠定基础。High performance liquid chromatography(HPLC)was used to detect the contents of various endogenous hormones from dry tuber to imbibed tuber to initially germinated tuber in order to reveal the change rule and regulation of endogenous hormones in Cyperus esculentus tuber.The results showed that the contents of abscisic acid(ABA),indoleacetic acid(IAA)and jasmonic acid(JA)were higher in the dormant dry tuber of Cyperus esculentus,and then exhibitted a trend of decreasing first and then increasing.On the contrary,the contents of gibberellin(GA 3)and zeatin riboside(ZR)were lower at the beginning,and then exhibitted a trend of increasing first and then decreasing,which indicated that ABA,IAA and JA were a kind of hormones inhibiting germination,whereas GA 3 and ZR were a kind of hormones promoting germination.Furthermore,their effects were verified by corresponding exogenous hormone treatments.On this basis,further study on the synergistic pathway between various hormones would lay a foundation for analyzing the regulation mechanism of dormancy and germination of Cyperus esculentus tuber.
