摘 要:目的探究谷胱甘肽过氧化物酶4(GPX4)、叉头框蛋白P2(FOXP2)在乳腺癌组织中的表达及其预后价值。方法选取2018年3月至2020年6月期间秦皇岛市第一医院收治的108例乳腺癌患者作为研究对象,收集术中切除的乳腺癌组织及癌旁组织标本,同时收集整理其临床资料。采用免疫组织化学法检测GPX4、FOXP2蛋白表达;采用Kaplan-Meier法分析乳腺癌组织中GPX4、FOXP2表达与患者预后的关系;Logistic回归分析乳腺癌患者预后的影响因素。结果与癌旁组织相比,乳腺癌组织中GPX4阳性表达率较高,FOXP2阳性表达率较低(P<0.05)。肿瘤直径≥3 cm、分化程度较低、TNM分期较晚、发生淋巴结转移的乳腺癌患者GPX4阳性表达、FOXP2阴性表达比例高于肿瘤直径<3 cm、分化程度较高、TNM分期较早、未发生淋巴结转移的患者(P<0.05)。乳腺癌组织GPX4阳性表达、FOXP2阴性表达的乳腺癌患者2年生存率低于乳腺癌组织GPX4阴性表达、FOXP2阳性表达的患者(P<0.05)。Logistic分析结果显示TNM分期、分化程度以及乳腺癌组织中GPX4、FOXP2表达情况均是乳腺癌患者预后的影响因素(P<0.05)。结论乳腺癌组织中GPX4呈高表达,FOXP2呈低表达,二者均是乳腺癌患者预后的影响因素。Objective To investigate the expression of glutathione peroxidase 4(GPX4)and forkhead box P2(FOXP2)in breast cancer and their prognostic value.Methods A total of 108 breast cancer patients admitted to First Hospital of Qinhuangdao from March 2018 to June 2020 were selected,specimens of breast cancer tissue and adjacent tissue removed during operation were collected,meantime,the clinical data were collected.The expression of GPX4 and FOXP2 proteins was detected by immunohistochemistry.Kaplan-Meier method was applied to analyze the relationship between the expression of GPX4,FOXP2 and the prognosis of breast cancer;and Logistic regression was applied to analyze the prognostic factors of breast cancer patients.Results Compared with the adjacent tissues,the positive expression rate of GPX4 was higher in breast cancer tissues,and the positive expression rate of FOXP2 was lower(P<0.05).The proportions of GPX4 positive expression and FOXP2 negative expression in breast cancer patients with tumor diameter≥3 cm,low degree of differentiation,late TNM stage and lymph node metastasis were higher than those in patients with tumor diameter<3 cm,high degree of differentiation,early TNM stage and no lymph node metastasis(P<0.05).The 2-year survival rate of breast cancer patients with GPX4 positive expression and FOXP2 negative expression was lower than that of breast cancer patients with GPX4 negative expression and FOXP2 positive expression(P<0.05).Logistic analysis showed that TNM stage,degree of differentiation and the expression of GPX4 and FOXP2 in breast cancer tissue were the prognostic factors of breast cancer patients(P<0.05).Conclusion GPX4 is overexpressed and FOXP2 is overexpressed in breast cancer,both of them are prognostic factors of breastcancerpatients.
