CKLF1过表达对肾癌细胞增殖、迁移及侵袭能力的影响及机制研究

摘　　要：肾癌是泌尿系统常见的恶性肿瘤.趋化素样因子1(chemokine-like factor 1,CKLF1)在多种恶性肿瘤中呈高表达,本研究旨在探讨CKLF1在肾癌细胞ACHN中过表达后,肾癌细胞ACHN增殖、迁移和侵袭能力的变化及其发生机制.利用4D核转染技术将实验组和对照组质粒分别转染肾癌ACHN细胞,实现该因子在肾癌细胞中的过表达.显微镜下,通过观察绿色荧光蛋白在视野细胞中的表达比例判定转染效率.利用Incu Cyte S3活细胞动态成像与分析系统评估细胞的增殖情况.划痕和transwell实验被用于评估CKLF1过表达后,肾癌细胞迁移和侵袭能力的变化情况.Western blot法检测CKLF1过表达后,蛋白CyclinD1、MMP2表达情况.动态细胞监测系统检测结果显示,与对照组相比较,实验组细胞的增殖活性明显高于对照组.CKLF1过表达后,实验组肾癌细胞的迁移和侵袭能力均明显升高,并且CyclinD1和MMP2蛋白表达水平明显高于对照组.CKLF1可能通过促进CyclinD1的表达增强细胞的增殖活性,通过促进MMP2蛋白的表达,增强ACHN细胞的迁移和侵袭能力.在肾癌的发生以及病程进展中CKLF1可能是重要的促癌因子.Renal carcinoma is a common malignant tumor of the urinary system.Chemokine-like factor 1(CKLF1)is highly expressed in a variety of malignant tumors.The aim of this study was to investigate the effects and mechanism of CKLF1 overexpression on the proliferation,migration,and invasion of renal cancer ACHN cells.4D nuclear transfection technique was used to transfect the plasmids of the experimental group and the control group into ACHN cells of renal carcinoma respectively to achieve overexpression of the factor in renal carcinoma cells.Under the microscope,the transfection efficiency was determined by observing the expression ratio of green fluorescent protein in the visual field cells.The Incu Cyte S3 Live cell dynamic imaging and analysis system was used to evaluate cell proliferation.Scratch and transwell assays were used to evaluate the changes in the migration and invasion ability of renal cell carcinoma after CKLF1 overexpression.Western blot was used to detect the expression of CyclinD1,MMP2 after overexpression of CKLF1.The results of the dynamic cell monitoring system showed that the proliferative activity of the experimental group was significantly higher than that of the control group.After CKLF1 overexpression,the migration and invasion ability of renal carcinoma cells in the experimental group was significantly increased,and CyclinD1 and MMP2 protein expression levels were significantly higher than those in the control group.CKLF1 can enhance the proliferation activity of ACHN cells by promoting the expression of CyclinD1,and enhance the migration and invasion ability of ACHN cells by promoting the expression of MMP2 protein.CKLF1 may be an important oncogenic factor in the development and progression of renal carcinoma.
