摘 要:目的:探究葛根素对产后母鼠泌乳作用和乳品质的影响及其可能机制。方法:将雌、雄KM小鼠共27只以3∶1比例合笼配种,得到孕鼠饲养至分娩。分娩后的母鼠随机分为正常对照组、葛根素低剂量组(葛根素溶液18 mg/kg)、葛根素高剂量组(葛根素溶液72 mg/kg),每组8只。从产后第3天起,每天灌胃1次,共10 d。观察实验周期内母鼠、仔鼠体质量变化,ELISA法检测乳腺组织中催乳素(PRL)含量,HE染色观察乳腺组织病理学形态,免疫组化法检测乳腺组织中催乳素受体(PRLR)、β-酪蛋白(β-casein)表达,Western Blot法检测乳腺组织中β-casein、葡萄糖转运蛋白1(GLUT1)、脂肪酸合成酶(FAS)的表达。结果:与正常对照组比较,葛根素低、高剂量组显著增加仔鼠体质量,母鼠体质量减少显著降低(P<0.05);葛根素低、高剂量组均可见乳腺小叶内腺泡变大,分泌物增多,且葛根素低剂量组更为显著;葛根素低、高剂量组乳腺中PRL含量显著增多(P<0.05,P<0.01);葛根素低剂量组β-casein、FAS的蛋白表达显著增加(P<0.05),而葛根素高剂量组PRLR、β-casein、FAS的蛋白表达显著降低(P<0.01)。结论:葛根素低剂量对产后母鼠有促进泌乳作用,且能够增强乳蛋白和乳脂相关蛋白表达,提升乳品质;葛根素在高剂量时促进泌乳作用不显著,且对乳品质有一定程度降低,可能与PRL浓度过高导致乳腺中PRLR蛋白表达受到抑制有关。Objective:To explore the effect of puerarin on lactation and milk quality in postpartum normal mice and its possible mechanism.Methods:Female and male KM mice were bred together in a 3:1 ratio,and the pregnant mice were bred until delivery.Mice after delivery were randomly divided into normal control group,low-dose puerarin(18 mg/kg of puerarin solution),and high-dose puerarin group(72 mg/kg of puerarin solution),with 8 mice in each group.From the 3rd day postpartum onwards,gavage was given once a day for a total of 10 days.The changes in body weight of mice and pups during the experimental period were observed,the content of prolactin(PRL)in breast tissue was detected by ELISA,the pathological morphology of breast tissue was observed by HE staining,and the prolactin receptor(PRLR)and β-casein in breast tissue were detected by immunohistochemistry.Western Blot method was used to detect the expression of β-casein,glucose transporter 1(GLUT1)and fatty acid synthase(FAS)in breast.Results:Compared with the normal control group,the low-dose and high-dose groups of puerarin significantly increased the body weight of pups,and decreased the body weight of mice(P<0.05),secretions increased significantly,and the low dose was more significant;the protein expressions of β-casein,and FAS in the low-dose puerarin group were significantly increased(P<0.01),while the PRLR,β-casein and FAS in the high-dose puerarin group were significantly increased(P<0.01).The protein expression was significantly decreased(P<0.01).Conclusion:Low dose of puerarin can promote lactation in postpartum mice,and can enhance the expression of milk protein and milk fat-related proteins,and improve milk quality;puerarin at high dose has no significant effect on promoting lactation,and decrease the milk quality to a certain extent,which might be related with high content of PRL induced suppression of PRLR protein expression in mammary gland.
