摘 要:目的 分析miR-150-5p对脑小血管病(CSVD)大鼠脑微血管内皮细胞凋亡的影响。方法 56只雄性大鼠14只作为正常组并不做处理,剩余42只建立CSVD模型,建模中途死亡6只。将建模成功的大鼠随机分为模型组、上调组、下调组,每组8只,做miR-150-5p转染,鉴定miR-150-5p转染率、氧化应激指标水平、神经凋亡率和乳酸脱氢酶(LDH)释放率。结果 与正常组、模型组相比,上调组miR-150-5p表达量升高,下调组miR-150-5p表达量降低,具有统计学差异(P<0.05),证明miR-150-5p转染成功。与正常组相比,模型组、下调组细胞凋亡率、LDH释放率、丙二醛(MDA)水平升高,超氧化物歧化酶(SOD)水平降低,上调组细胞凋亡率、LDH释放率、MDA水平降低,SOD水平升高,具有统计学差异(P<0.05);与模型组相比,下调组细胞凋亡率、LDH释放率、MDA水平升高,SOD水平降低,上调组细胞凋亡率、LDH释放率、MDA水平降低,SOD水平升高,具有统计学差异(P<0.05);与上调组相比,下调组细胞凋亡率、LDH释放率、MDA水平升高,SOD水平降低,具有统计学差异(P<0.05)。结论 miR-150-5p可能作用于磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(Akt)信号通路,影响CSVD大鼠脑微血管内皮细胞凋亡。Objective To analyze the effect of miR-150-5p on apoptosis of cerebral microvascular endothelial cells in rats with cerebral small vessel disease(CSVD).Methods 14 of 56 male rats served as the normal group and were not treated.The remaining 42 rats were used to establish CSVD models,and 6 rats died midway through the modeling process.The successfully modeled rats were randomly divided into model group,up-regulated group,and down-regulated group,with 8 rats in each group.miR-150-5p transfection was performed to identify the miR-150-5p transfection rate,oxidative stress index levels,neuronal apoptosis rate,and lactic dehydrogenase(LDH)release rate.Results Compared with the normal group and model group,the expression of miR-150-5p in the up-regulated group was increased,the expression of miR-150-5p in the down-regulated group was decreased,with a statistically significant difference(P<0.05),indicating successful transfection of miR-150-5p.Compared with normal group,apoptosis rate,LDH release rate and malondialdehyde(MDA)level were increased,and superoxide dismutase(SOD)level was decreased in model group and down-regulated group,while apoptosis rate,LDH release rate and MDA level were decreased,and SOD level was increased in up-regulated group,with a statistically significant difference(P<0.05).Compared with model group,apoptosis rate,LDH release rate and MDA level were increased and SOD level was decreased in down-regulated group,while apoptosis rate,LDH release rate and MDA level were decreased and SOD level was increased in up-regulated group,with a statistically significant difference(P<0.05).Compared with the up-regulated group,the apoptosis rate,LDH release rate,MDA level were increased and SOD level was decreased in the down-regulated group,with a statistically significant difference(P<0.05).Conclusions miR-150-5p might act on the phosphatidylinositol 3-kinase(PI3K)/protein kinase B(Akt)signaling pathway,affecting the apoptosis of cerebral microvascular endothelial cells in CSVD rats.
