摘 要:为了获得拟无枝酸菌QT-25(Amycolatopsis sp.QT-25)的基因组序列信息,本研究通过分析其次级代谢产物生物合成基因簇预测其代谢产物,为发现潜在的新结构抗生素提供科学依据。基于16S rRNA基因序列进行菌株属水平鉴定,利用Illumina HiSeq+PacBio测序技术对拟无枝酸菌QT-25进行基因组测序,采用antiSMASH(v6.0.1)在线工具分析次级代谢产物生物合成基因簇。结果表明,拟无枝酸菌QT-25基因组序列全长8010145 bp,平均G+C含量为68.66%,含有共编码7488个基因,预测到该菌株至少含有30个次级代谢产物生物合成基因簇,其中Cluster 4具有编码糖肽类抗生素的潜力。研究结果揭示,来自贺兰山岩石地衣的拟无枝酸菌QT-25具有丰富的次级代谢产物产生能力,有进一步发掘新结构抗生素的价值。In order to obtain the genome sequence information of Amycolatopsis sp.QT-25,this study predicted its metabolites by ana-lyzing the biosynthetic gene clusters of secondary metabolites,which would provide a scientific basis for the discovery of potential new structural antibiotics.The strain Amycolatopsis sp.QT-25 was identified at the genus level based on the 16S rRNA gene sequence and the genome of Amycolatopsis sp.QT-25 was sequenced by Illumina HiSeq+PacBio sequencing technology.Furthermore,its secondary metabolite biosynthetic gene clusters were analyzed by antiSMASH(v6.0.1)online tool.The results showed that the genome sequence of Amycolatopsis sp.QT-25 was 8010145 bp in length,with an average G+C content of 68.66%,containing 7488 genes.It was pre-dicted that the strain had at least 30 secondary metabolite biosynthetic gene clusters,among which cluster 4 had the potential to encode glycopeptide antibiotics.The results in this study demonstrated that Amycolatopsis sp.QT-25 from Helan Mountain rock lichens has a rich ability of secondary metabolite production,which has the value of further exploring new structural antibiotics.
