摘 要:目的探讨鼻咽癌(NPC)组织中BMI1和ERK3的表达、两者的相关性及其在NPC侵袭转移中的作用。方法检索并下载GEO数据库中NPC测序基因集GSE103611和GSE34573,分析BMI1和ERK3基因表达数据。采用免疫组化MaxVision法检测51例NPC患者组织和18例慢性鼻咽炎组织中BMI1和ERK3的表达情况,结合随访资料分析以上指标的临床意义。结果BMI1在NPC组织和慢性鼻咽炎组织中的阳性率分别是56.9%和22.2%,ERK3在NPC组织和慢性鼻咽炎组织中的阳性率分别是52.9%和11.1%。BMI1和ERK3在NPC组织中的阳性率均高于慢性鼻咽炎组织(P<0.05),尤其在NPC的N2~3组中两者呈显著高表达。BMI1与ERK3的表达呈正相关(r_(s)=0.345,P<0.05)。同时在NPC组织中BMI1和ERK3的表达与T分期、N分期以及总分期有关(P<0.05),与患者性别、初诊血清EBV DNA拷贝数无关(P>0.05)。结论BMI1/ERK3通路可在NPC的侵袭转移中发挥一定的作用。Objective To investigate the expression and correlation between BMI1 and ERK3 in nasopharyngeal carcinoma(NPC)tissues,and their roles in the invasive metastasis of NPC.Methods The NPC sequence gene sets GSE103611 and GSE34573 in the GEO database were retrieved and downloaded to analyze BMI1 and ERK3 gene expression.And the expression of BMI1 and ERK3 in the tissues of 51 patients with NPC and 18 patients with chronic rhinopharyngitis were assessed by the immunohistochemical in MaxVision method,and the clinical value of the above indexes was analyzed together with the follow-up data.Results The positive rates of BMI1 in NPC tissues and chronic nasopharyngitis tissues were 56.9%and 22.2%,respectively;while the positive rates of ERK3 in NPC tissues and chronic nasopharyngitis tissues were 52.9%and 11.1%,respectively.Both BMI1 and ERK3 were significantly more positive in NPC tissues than in chronic nasopharyngitis tissues(P<0.05),and both were significantly higher expressed in the NPC N2-3 group particularly.The expression of BMI1 and ERK3 had a significant positive correlation(r_(s)=0.458,P<0.05).Meanwhile,the expression of BMI1 and ERK3 in NPC tissues was correlated with T-stage,N-stage,and total stage(P<0.05),but not with patient gender,or initial serum EBV DNA copy number(P>0.05).Conclusion The BMI1/ERK3 pathway may play roles in the invasive metastasis of NPC.
