摘  要:旨在探究促性腺激素抑制激素(GnIH)对瑶鸡卵巢糖代谢的影响。将10只瑶鸡随机分为对照组(0.9%生理盐水)和GnIH组(24.54μg/kg GnIH),每组5只。每天7:00和19:00注射生理盐水或GnIH(100μL/次),连续注射14 d,观察并记录瑶鸡的采食情况;采用实时荧光定量PCR方法对瑶鸡卵巢中糖代谢相关基因葡萄糖转运蛋白1(GLUT1)、葡萄糖转运蛋白3(GLUT3)、丙酮酸激酶(PK)、磷酸果糖激酶(PFK)、乳酸脱氢酶A(LDHA)、乳酸脱氢酶B(LDHB)和琥珀酸脱氢酶B(SDHB)mRNA表达量进行检测;采用试剂盒对卵巢中葡萄糖、丙酮酸含量进行检测。结果:与对照组相比,GnIH组注射后1、2、4、8 h,瑶鸡的采食量显著升高(P<0.05),夜间平均采食量极显著升高(P<0.01),卵体比显著下降(P<0.05);GLUT1、GLUT3 mRNA的表达量极显著上升(P<0.01),PK、PFK和LDHA mRNA的表达量显著上升(P<0.05),LDHB和SDHB mRNA的表达量显著下降(P<0.05);丙酮酸含量显著升高(P<0.05)。本试验结果表明,腹腔注射GnIH会增加雌性瑶鸡采食量,引起卵巢糖代谢紊乱,进而导致卵巢发育异常。To determine the effect of gonadotropin-inhibiting hormone(GnIH)on ovarian glucose metabolism in Yao chickens,10 Yao chickens were obtained and randomly divided into the control group(treated with 0.9%saline)and the GnIH group(treated with 24.54μg/kg GnIH),with 5 birds in each group.Saline or GnIH(100μL/time)was injected daily at 7:00 and 19:00 for 14 days.Then,the feeding of these Yao chickens were observed and recorded.The mRNA expression of glucose metabolism-related genes glucose transporter protein 1(GLUT1),glucose transporter protein 3(GLUT3),pyruvate kinase(PK),phosphofructokinase(PFK),lactate dehydrogenase A(LDHA),lactate dehydrogenase B(LDHB)and succinate dehydrogenase B(SDHB)in the ovaries of the Yao chickens was examined by real-time fluorescence quantitative PCR.The glucose and pyruvate levels in the ovaries were measured using a completed kit.The results were:Compared with the control group,the feed intake of yoyos in the GnIH group was significantly higher at 1 h,2 h,4 h and 8 h after injection(P<0.05),and their average feed intake at night was highly significantly higher(P<0.01),and their egg-to-body ratio was significantly lower(P<0.05);the expressions of GLUT1 and GLUT3 mRNA in the tested birds were highly significantly higher(P<0.01).The expressions of PK,PFK and LDHA mRNAs in them were significantly increased(P<0.05).However,the expressions of LDHB and SDHBmRNA were significantly decreased(P<0.05);the pyruvate content was significantly increased(P<0.05).To conclude,intraperitoneal injection of Gn IH resulted in an increase in food intake in the female Yao chickens,causing disturbances in the ovarian glucose metabolism in them,which in turn led to their abnormal ovarian development.
