摘　　要：为探讨马铃薯开放式组织培养中次氯酸钠浸泡容器消毒浓度、培养基中抑菌剂浓度、蔗糖浓度、琼脂浓度对脱毒苗污染率、有效生长率的影响,在单因素试验的基础上,使用Design-Expert软件进行Box-Behnken响应面优化设计对马铃薯开放式组织培养条件进行优化,选取有效生长率为指标建立回归方程得到预测组合,综合考虑实际操作因素,对预测结果进行修正,验证修正结果得到最佳组合。结果表明,各因素对有效生长率的影响为次氯酸钠>蔗糖>益培隆>琼脂;修正得到开放式组织培养最优组合为次氯酸钠浸泡容器消毒浓度1%、培养基益培隆浓度0.5 mL/L、蔗糖浓度7.5 g/L、琼脂浓度6.6 g/L,经验证脱毒苗有效生长率可达85%,与预测结果相近。In order to investigate the influence of disinfection concentration of sodium hypochlorite immersion container,bacteriostatic concentration,sucrose concentration and agar concentration on contamination rate and effective growth rate of virus-free seedlings in potato open tissue culture.On the basis of single factor experiment,Box-Behnken response surface optimization design was carried out with Design-Expert software to optimize the conditions of potato open tissue culture.The effective growth rate was selected as the index to establish the regression equation to obtain the prediction combination.Taking the actual operation factors into consideration,the prediction results were modified to verify the optimal combination of the modified results.The results showed that the order of influence of each factor on effective growth rate was sodium hypochlorite>sucrose>Iperon>agar.The optimal combination of open tissue culture was modified as follows:sodium hypochlorite immersion container disinfection concentration of 1%,medium Yiperon concentration of 0.5 mL/L,sucrose concentration of 7.5 g/L,agar concentration of 6.6 g/L,and the effective growth rate of virulent seedlings proved to be 85%,which was close to the predicted result.
