摘 要:通过对黄牛乳进行乳成分分析及TMT定量蛋白质组学分析,筛选不同海拔黄牛乳成分差异表达蛋白,结合DAVID数据库分别进行乳清蛋白、乳脂肪球膜蛋白(MFGM)的GO和KEGG富集分析,使用STRING在线工具和Cytoscape 3.8.2软件构建不同海拔地区黄牛乳清、MFGM蛋白的蛋白质互作网络。结果,两种海拔中,3 850 m海拔地区(后记作HA)黄牛乳营养成分高于2 700 m海拔地区(后记作LA)黄牛乳;HA黄牛乳清蛋白、MFGM蛋白浓度显著高于LA黄牛乳清蛋白与MFGM蛋白。乳清蛋白中123个差异表达蛋白上调,122个差异表达蛋白下调;MFGM蛋白中172个差异表达蛋白上调,95个差异表达蛋白下调。黄牛乳清蛋白、MFGM蛋白共同参与免疫调控过程、代谢过程、发育过程;共同涉及的分子功能包括调节酶活性、能量结合、脂质结合。补体和凝血级联、溶酶体代谢、Ig A产生的肠道免疫网络、抗原加工和呈递、B细胞受体信号通路、NOD样受体信号通路、自然杀伤细胞介导的细胞毒性是主要的免疫相关信号通路。结果表明,HA黄牛乳的蛋白浓度更高,可使犊牛血清总蛋白(STP)升高,从而可影响犊牛成年后的繁殖性能。HA黄牛乳清、MFGM蛋白可能在抑制肿瘤发生、增强宿主免疫防御的信号通路中发挥重要作用,LA黄牛乳汁在抑制炎症反应过程中发挥作用。In this study,TMT proteomics technology was used to analyze the differential proteomics of whey protein and fat globule membrane protein of yellow cattle at different altitudes in Gannan area,which laid a foundation for understanding the protein level differences and related molecular mechanisms of yellow milk at different altitudes,and also provided a channel for the optimization of yellow cattle germplasm resources.The differentially expressed proteins of milk components in yellow cattle at different altitudes were obtained by TMT quantitative proteomics analysis.Combined with DAVID database,GO and KEGG enrichment analysis of whey protein and milk fat globule membrane protein(MFGM protein)were obtained respectively.Cytoscape 3.8.2 software was used to construct protein interaction network analysis of whey and MFGM proteins in yellow cattle at different altitudes.In result,the concentration of whey protein and MFGM protein in relatively high altitude(3850 m)was signif icantly higher than that in relatively low altitude(2700 m).123 differentially expressed proteins were up-regulated and 122 differentially expressed proteins were down-regulated.In MFGM protein,172 differentially expressed proteins were up-regulated and 95 differentially expressed proteins were down-regulated.The biological processes involved in whey protein and MFGM protein in cattle at different altitudes mainly included immune regulation process,metabolic process and development process.The molecular functions involved include regulating enzyme activity,energy binding,lipid binding,etc.Common cellular components include extracellular regions,membrane-bound organelles,and extracellular bodies.The complement and coagulation cascades,lysosomal metabolism,intestinal immune network produced by IgA,antigen processing and presentation,B cell receptor signaling pathway,NOD-like receptor signaling pathway,and natural killer cell-mediated cytotoxicity were the main immune-related signaling pathways.The above results showed that the milk protein concentrat
