摘  要:甘油-3-磷酸酰基转移酶(GPAT)是一类催化甘油酯质合成的膜结合酶,可用于多种植物的基因家族鉴定和分析,并在植物生长发育和逆境中发挥着重要作用。针对四倍体小麦的GPAT基因,对其进行了家族鉴定、基因结构、进化关系等比较分析。结果表明:在硬粒小麦和野生二粒小麦中各鉴定出33个GPAT家族成员,分别命名为TtGPAT1~TtGPAT33、TdGPAT1~TdGPAT33,并对其基因结构、保守基序、系统进化、共线性分析时发现,TtGPATs和TdGPATs蛋白可以分为GroupⅠ、GroupⅡ、GroupⅢ等3类,且在同一分组中,TtGPATs和TdGPATs成员数量几乎一致,大部分GroupⅠ和GroupⅡ成员有7~12个内含子,GroupⅢ成员有1~3个内含子;其蛋白质的理化性质分析表明:GPAT蛋白成员的氨基酸长度为149(TdGPAT19)~589(TtGPAT1),分子量为16.22 kDa(TdGPAT19)~62.76 kDa(TtGPAT1),等电点为5.92(TdGPAT5)~11.39(TdGPAT7);硬粒小麦与拟南芥的GPAT基因之间仅存在2个共线性关系,但与野生二粒小麦存在66个共线性基因对,这为四倍体小麦基因功能的研究提供了参考,也对拓宽六倍体小麦的遗传基础具有重要的理论和应用价值。Glycerol-3-phosphateacyltransferase(GPAT),a class of membrane-bound enzymes that catalyze glycerolipid synthesis,was analyzed for gene family identification in a variety of plants and plays an important role in plant growth and development and adversity.In this study,a comparative analysis of family identification,gene structure,and evolutionary relationships was performed for GPAT genes in tetraploid wheat.The results showed that 33 GPAT family members,named TtGPAT1~TtGPAT33 and TdGPAT1~TdGPAT33,were identified in each of durum wheat and wild dicot wheat,and their gene structure,conserved motifs,phylogenetic and covariance analyses revealed that TtGPATs and TdGPATs proteins can be classified into GroupⅠ,GroupⅡand GroupⅢ,and the number of TtGPATs and TdGPATs members in the same group is almost the same,most of the GroupⅠand GroupⅡmembers have 7~12 introns,and GroupⅢmembers have 1~3 introns.Physicochemical property analysis shows that the amino acid length of GPAT members is 149(TdGPAT19)~589(TtGPAT1),molecular weight 16.22 kDa(TdGPAT19)~62.76 kDa(TtGPAT1),and isoelectric point 5.92(TdGPAT5)~11.39(TdGPAT7).There were only 2 covariates between durum wheat and Arabidopsis GPAT genes,but,but 66 covariate gene pairs existed with wild two-grain wheat.These results provide a reference for the study of gene function in tetraploid wheat,and also have important theoretical and application implications for broadening the genetic basis of hexaploid wheat.
