摘　　要：阿尔茨海默病(AD)起病隐匿,发病机制尚不清楚,脑内β-淀粉样蛋白(Aβ)斑块是AD的主要病理学特征。仑卡奈单抗是一种靶向Aβ的单克隆抗体药物,2023年7月6日获得美国食品药品监督管理局完全批准用于早期AD的治疗。仑卡奈单抗主要通过与可溶性Aβ分子结合形成免疫复合物,促进患者脑内Aβ斑块的清除。临床Ⅲ期研究结果表明,仑卡奈单抗使患者认知功能的下降速度减缓27%,临床应用中患者耐受性良好,最常见的不良反应为输注相关反应。目前,该药尚未在我国获批上市,本文对仑卡奈单抗的临床研究进展进行简要综述,以期为AD临床治疗提供一定的用药参考和理论依据。Alzheimer's disease(AD)has an insidious onset and unclear pathogenesis,amyloidβ-protein(Aβ)plaque in the brain is the main pathologic feature of AD.Lencanumab is a monoclonal antibody targeting Aβ,received full approval of FDA on July 6,2023 for the treatment of early AD.Lencanumab mainly promotes the clearance of Aβplaques by binding with soluble Aβmolecules to form immune complexes.The results of the phaseⅢclinical study showed that lencanumab was able to slow down the decline rate of cognitive dysfunction by 27%,and it was well tolerated by patients in clinical applications,and the most common adverse reactions are infusion-related reactions.At present,this drug has not been approved in China.This article provides a brief review of the clinical research progress of lencanumab,with a view to providing a certain drug reference and theoretical basis for the clinical treatment of AD.
