摘　　要：目的研究神经酰胺在AngⅡ诱导人脐静脉内皮细胞凋亡过程中的作用。方法体外培养人脐静脉内皮细胞,采用不同浓度的AngⅡ和FB1干预血管内皮细胞,采用流式细胞术检测细胞凋亡情况。结果AngⅡ处理组血管内皮细胞的阳性凋亡率显著高于对照组(P<0.05)且具有时间和剂量依赖性,对照组和终浓度分别为10^(-7)mol/L、10^(-6)mol/L、10^(-5)mol/L的AngⅡ处理血管内皮细胞24h后各组细胞凋亡率分别为(2.3±1.0)%、(6.2±1.1)%、(16.9±2.2)%、(28.8±4.4)%;终浓度为10^(-6)mol/L的AngⅡ处理血管内皮细胞6 h、24 h、48 h后细胞凋亡率分别为(6.4±2.4)%、(14.4±3.3)%、(22.7±2.8)%;FB1处理组结果显示血管内皮细胞凋亡率显著降低(P<0.05)。结论AngⅡ能诱导细胞凋亡,神经酰胺可能是其途径之一。Objective To explore the role of ceramide in the apoptosis of human umbilical vein endothelial cells(EC)induced by AngiotensinⅡ(AngⅡ).Methods Human umbilical endothelial cells were primarily cultured in vitro and then they were intervened with different doses of AngⅡand FB1.Flow cytometry was used to detect the apoptosis of EC.Results The positive rate of EC in AngⅡ-treated group was significantly higher than that in control group(P<0.05)in a time-and dose-dependent manner.The rate of the apoptotic EC were(2.3±1.0)%,(6.2±1.1)%,(16.9±2.2)%and(28.8±4.4)%,respectively after treated with AngⅡat concentrations of 0,10^(-7),10^(-6),10^(-5) mol/L for 24 hours.After treatment with AngⅡ(10^(-6) mol/L)for 6h,12h and 24h,the rate of apoptotic cells were(6.4±2.4)%,(14.4±3.3)%and(22.7±2.8)%respectively.FB1 treatment results showed that the apoptosis of EC was significantly reduced(P<0.05).Conclusions AngⅡcan induced apoptosis of endothelial cells,and ceramide may be one of its pathway.
