摘 要:目的:探讨沉默色素框同源蛋白4(CBX4)对食管胃腺癌细胞增殖、迁移和侵袭能力的影响。方法:RT-qPCR检测2017年6月至2019年6月于山西省肿瘤医院进行食管胃结合部腺癌手术患者的癌组织和癌旁组织及食管胃结合部腺癌细胞系中CBX4的mRNA表达水平。以食管胃结合部腺癌细胞AGS和SK-GT-4为研究对象,分别转染阴性对照(NC)和CBX4 si RNA,采用CCK-8试剂盒检测细胞增殖,划痕实验检测细胞迁移,Transwell实验检测细胞的侵袭,Western blot检测细胞中CBX4、β-catenin、c-MYC和cyclin D1的蛋白表达水平。结果:(1)与临近正常组织相比,食管胃结合部腺癌组织中CBX4表达量明显上调;(2)与正常胃上皮细胞GES-1相比,食管胃结合部腺癌细胞中CBX4 mRNA水平显著升高(P<0.05);(3)沉默CBX4表达后,AGS和SK-GT-4细胞增殖、迁移和侵袭能力明显下降;(4)沉默CBX4后,Wnt/β-catenin信号通路相关蛋白表达均降低(P<0.05)。结论:CBX4在食管胃结合部腺癌组织和细胞中高表达,沉默CBX4可能通过调控Wnt/β-catenin信号通路抑制食管胃结合部腺癌细胞的增殖、迁移和侵袭。AIM:To explore the effect of chromobox 4(CBX4)silencing on the proliferation,migration and invasion of esophageal and gastric junction adenocarcinoma cells.METHODS:RT-qPCR was used to detect the expres‐sion level of CBX4 mRNA in cancer tissues and adjacent tissues of patients with esophageal and gastric junction adenocar‐cinoma who underwent surgery in Shanxi Provincial Cancer Hospital from June 2017 to June 2019,and in esophageal and gastric junction adenocarcinoma cell lines.Taking the esophageal and gastric junction adenocarcinoma cells AGS and SK-GT-4 as the research objects,negative control(NC)and CBX4 siRNAs were transfected into the cells.Cell proliferation was detected by CCK-8 assay,cell migration was detected by scratch test,and cell invasion was detected by Transwell test.Western blot was used to detect CBX4,β-catenin,c-MYC and cyclin D1 protein expression levels.RESULTS:Compared with adjacent normal tissues,the expression of CBX4 in esophageal and gastric junction adenocarcinoma was significantly up-regulated.Compared with normal gastric epithelial cells,the level of CBX4 mRNA in esophageal and gas‐tric junction adenocarcinoma cells was significantly increased(P<0.05).After down-regulation of CBX4 expression,the proliferation,migration and invasion of AGS and SK-GT-4 cells decreased significantly,and the expression ofβ-catenin,c-MYC and cyclin D1 protein also decreased(P<0.05).CONCLUSION:CBX4 is highly expressed in esophageal and gastric junction adenocarcinoma cells.Silencing of CBX4 may regulate Wnt/β-catenin signaling pathway to inhibit the pro‐liferation,migration and invasion of esophageal and gastric junction adenocarcinoma cells.
