摘  要:目的 利用在线生物信息学数据库,通过组织和细胞水平的实验验证CapG基因在肝癌中的表达和临床意义。方法通过分析UALCAN和GEPIA数据库,获得CapG基因在肝细胞癌患者中的mRNA转录表达水平、DNA甲基化状态和临床病理特征等;用Kaplan-Meier Plotter绘制肝细胞癌患者的生存曲线,用HPA分析CapG的蛋白水平,用Kaplan-Meier Plotter绘制肝细胞癌患者的生存曲线,用HPA分析CapG的蛋白水平。用String网站构建相应蛋白的相互作用网络,用LinkedOmics工具对CapG共表达的基因进行富集分析;此外,用免疫组化法验证CapG在肝癌组织芯片中的表达,用定量PCR法测定CapG在肝癌细胞水平的表达。结果 肝癌患者肝组织中CapG基因的mRNA表达水平和蛋白水平均明显升高(P<0.001),CapG的高mRNA表达水平与DNA甲基化状态的降低明显相关。免疫组织学和细胞水平的实验结果证实了CapG在肝细胞癌中的明显高表达(P<0.05)。String在线分析结果表明,与CapG相互作用的蛋白质参与了生物过程,包括参与肌动蛋白聚合、转录调控、蛋白质运输等生物过程;LinkedOmics富集分析表明,这些共同表达的基因在肝癌组织中可以促进细胞因子代谢过程、T细胞激活、免疫系统的调节、细胞黏附调节以及细胞黏附的蛋白质复合物的合成等。此外,CapG mRNA的高表达水平与肝癌患者的总生存期和疾病特异性生存期显著相关(P<0.001)。结论 基于生物信息学和组织及细胞水平的实验表明,肝癌组织中CapG基因的高表达与肝癌患者的不良生存期密切相关。CapG基因可能是诊断和治疗肝癌患者的潜在靶点。Objective To explore the expression and clinical significance of CapG gene in hepatocellular carcinoma through bioin-formatics analysis based on tissues and cell tests.Methods UALCAN database and GEPIA database were utilized to analyze the mRNA transcription level,DNA methylation status and clinicopathological characteristics of CapG gene;Kaplan-Meier plotter was used to plot the survival curve of patients with liver cancer.HPA database and tissue chip were used to evaluate the protein expression levels of CapG in hepatocellular carcinoma patients.String database was applied to construct protein-protein interaction(PPI)network of CapG coding protein.LinkedOmics was used for function enrichment analysis of CapG.Furthermore,the protein level of CapG was detected by immunohistochemistry liver cancer tissue chips,and quantitative PCR was used to determine the mRNA level of CapG in different liver cancer cells.Results mRNA and protein levels of CapG gene in hepatocellular cancer tissues were significantly higher than those in normal hepatocellular tissues(P<0.001),while the high expression level of CapG mRNA was significantly correlated with the decrease of DNA methylation status.Immunohistochemical and cellular assay confirmed abnormally high expression of CapG in liver cancer(P<0.05),which was correlated with pathological grade of liver cancer patients(P<0.001).PPI network suggested that CapG protein was closely associated with biological processes,including actin polymerization,transcriptional regulation,protein transport and other biological processes;The enrichment analysis of LinkedOmics showed that CapG positively correlated genes involving in the process of cytokine metabolism,activation of T cells,regulation of cell adhesion,regulation of immune system and synthesis of protein complexes of cell adhesion.In addition,high expression of CapG mRNA was significantly associated with overall survival and disease-specific survival(P<0.001).Conclusion In this study,results based on bioinformatics database,as well as t
