摘 要:目的:优选雾水葛总黄酮涂膜剂成膜材料及其辅料的配比并检测涂膜剂的抑菌和抗氧化效果。方法:采用单因素试验和正交试验考察成膜材料PVA-124和白及胶浆、增塑剂甘油、乳化剂吐温-80、增强剂可溶性淀粉的用量对成膜性能的影响;纸片扩散法考察雾水葛涂膜剂对绿脓杆菌、金黄色葡萄球菌、大肠杆菌的抑菌作用;通过测定对DPPH自由基清除率和还原能力,考察雾水葛总黄酮涂膜剂的抗氧化能力。结果:最佳配比组合为A1B3C2D3E3,即PVA-124 5.0 g、白及胶浆4 g、甘油1.5 g、吐温-80 0.8 g和0.75 g可溶性淀粉;含50 mg/mL和100 mg/mL总黄酮的雾水葛总黄酮涂膜剂的抑菌圈与阳性对照组比,其抑菌效果不如阳性对照组;在0.05~0.8 mg/mL范围内,雾水葛总黄酮涂膜剂对DPPH自由基清除率随浓度升高而增大,最高可达(96.42±0.52)%,雾水葛总黄酮涂膜剂的还原能力随浓度升高而增大,最高可达(96.43±1.33)%。结论:该工艺确定的成膜材料及其辅料配比合理,成膜性能最优;雾水葛总黄酮涂膜剂有一定的抑菌能力和抗氧化能力,具有一定的抗炎祛痘功效。Objective:To optimize the ratio of film forming materials and auxiliary materials of Pouolia seylanic(L.) Benn coating agent and to detect the antibacterial and antioxidative effects of coating agents.Methods:Single factor test and orthogonal test were used to investigate the effect of film forming material PVA-124 and White enamel glue, plasticizer glycerin, emulsifier Tween-80,and enhancer soluble starch on film forming properties.Paper diffusion method to investigate the antibacterial activity of Pouolia seylanic(L.) Benn coating agent on Pseudomonas aeruginosa, Staphylococcus aureus and Escherichia coli The antioxidant capacity of Pouolia seylanic(L.) Benn coating agent was investigated by measuring the DPPH free radical scavenging rate and reducing ability.Results:The best combination was A_1B_3C_2D_3E_3,ie PVA-124 5.0g, white and glue 4 g, glycerol 1.5g, Tween-80 0.8g and 0.75g soluble starch;containing 50mg/mL and 100mg/mL total flavonoids The ratio of the inhibition zone of the Pouolia seylanic(L.) Benn coating agent to the positive control group was P≤0.01,and the ratio to the blank group was P≤0.01.The antibacterial effect was not as good as that of the positive control group;in the range of 0.05~0.8mg/mL,The DPPH free radical scavenging rate increased with the increase of concentration, and the highest can reach(96.42±0.52)%.The reducing ability of the Pouolia seylanic(L.) Benn coating agent increased with the increase of the concentration, the highest(96.43±1.33)%.Conclusion:The ratio of film-forming materials and their auxiliary materials determined by this process is reasonable, and the film-forming performance is optimal.Pouolia seylanic(L.) Benn coating agent has certain antibacterial and anti-oxidation ability, but the antibacterial ability is not as good as Antibiotic.
