摘 要:多酚是桑叶的主要活性成分之一,为了优化桑叶多酚的提取测定方法,并研究多酚在不同品种、不同采样时间间的变化规律,本研究利用单因素试验依次考察了粉碎粒度、提取溶剂、提取温度、提取时间对福林酚法测定桑叶多酚含量的影响,并测定了20个桑叶品种多酚含量在3个采样时间的变化以及与1-脱氧野尻霉素(Deoxynojirimycin,DNJ)的相关性。结果显示,桑叶多酚的最优提取条件为粉碎粒度为过80目筛、料液比1∶50、70%乙醇80℃水浴加热回流1 h,该方法具有较高的可重复性和准确性。利用该方法测定了20个桑品种在2022年4月下旬、5月上旬和8月上旬的多酚含量,发现了不同桑品种在该3个采样时间多酚含量总体逐渐降低的变化规律。将多酚含量与DNJ含量进行关联分析发现,桑叶多酚与DNJ呈负相关。Polyphenols are one of the main active components of mulberry leaves.In order to optimize the extraction and determination method of polyphenols from mulberry leaves,and to study the variation of polyphenols in different varieties at different sampling times,the effects of grinding particle size,extraction solvent,extraction temperature and extraction time on the determination of polyphenols in mulberry leaves were investigated by single factor experiment.The changes of polyphenol contents in 20 mulberry varieties at three sampling times and their correlation with 1-deoxynojirimycin(DNJ)were determined.The results showed that the optimal extraction conditions of mulberry leaf polyphenols were as follows:grinding particle size was 80 mesh sieve,solid-liquid ratio was 1∶50,and 70%ethanol was heated and reflux in 80℃water bath for 1 h.The method had high repeatability and accuracy.This method was used to determine the contents of polyphenols in 20 mulberry varieties in late April,early May and early August in 2022,and it was found that the contents of polyphenols in different mulberry varieties gradually decreased at these three sampling times.The correlation analysis showed that polyphenol content and DNJ content were negatively correlated.
