摘 要:非结构性碳水化合物是植物体内碳水化合物的一种重要存在形式,其含量高低与植物生长发育、逆境胁迫响应密切相关。为分析桑叶中非结构性碳水化合物含量,本研究利用80%乙醇、75%甲醇和MCW介导的不同抽提方法,设置不同条件进行测定对比分析。结果显示:75%甲醇和MCW抽提获得的可溶性糖产物无明显色素干扰,且75%甲醇方法抽提产物的可溶性糖与淀粉比值更高;增加抽提重复次数时,可溶性糖抽提得率增加。本研究经综合分析提出75%甲醇恒温振荡抽提1 h是适合桑叶非结构性碳水化合物的简便快速抽提方法。这为今后开展桑树生长发育的生理学以及桑树品种抗逆性能等分析提供方法学参考。As an important form of carbohydrates,non-structural carbohydrate plays key roles in growth,development and stress response in plant.In order to analyze the non-structural carbohydrate content in mulberry leaves,different extraction methods including methods mediated by 80%ethanol,75%methanol and MCW were compared in this study.Results showed that the soluble carbohydrate extracted by 75%methanol and MCW had relatively higher purity than that by 80%ethanol.The soluble sugar to starch ratio of the product extracted by 75%methanol was higher.The concentration of acquired soluble carbohydrate increased with the increasement of extraction repeats.Extraction with 75%methanol for 1 h is recommended as a simple and rapid method suitable for non-structural carbohydrates analysis in mulberry leaves.This present study provides a methodological reference for mulberry physiology research and variety selections in the future.
