摘 要:目的研究铁皮石斛(Dendrobium officinale Kimura et Migo)茎中的苯丙素类化学成分。方法对铁皮石斛茎的水提取物和体积分数95%乙醇提取物采用Diaion HP-20大孔吸附树脂柱色谱、硅胶柱色谱、ODS柱色谱、Sephadex LH-20柱色谱和高效液相色谱(HPLC)等技术进行分离纯化,根据波谱学数据鉴定各化合物的结构。结果从铁皮石斛茎的提取物中分离鉴定15个苯丙素类成分,分别为alangilignoside B(1)、(-)-(7S,8R)-4,9′-di-O-β-D-glucopyranosyloxydihydrodiconiferyl alcohol(2)、5,5′-二甲氧基落叶松脂素-4-O-β-D-葡萄糖苷(3)、icariol A2-4-O-β-D-glucoside(4)、picraquassioside C(5)、syringylgcerol-8-O-4′-(synapyl alcohol)ether(6)、(+)-lyoniresionl-9-O-β-D-glucopyranoside(7)、鹅掌楸苷(8)、丁香脂素-4-O-β-D葡萄糖苷(9)、N-反式香豆酰酪胺(10)、shashenoside(11)、丁香酚-O-β-D-芹糖-(1″→2′)-O-β-D-葡萄糖苷(12)、3-(4-羟基-3,5二甲氧基苯基)-丙烷-1,2-二醇(13)、二氢阿魏酸(14)和松柏苷(15)。化合物4和7显示弱的2,2′-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)自由基清除能力,化合物4、7和12具有中等程度的2,2-二苯基-1-若基肼(DPPH)自由基清除能力。结论化合物1、2、7、11~14为首次报道从石斛属植物中分离得到,化合物3、5、6和8为首次报道从铁皮石斛中分离得到。化合物4、7和12具有一定的抗氧化活性。OBJECTIVE To study the phenylpropanoids from the stems of Dendrobium officinale Kimura et Migo.METHODS The water extract and 95%ethanol extract from the stems of Dendrobium officinale were isolated and purified by Diaion HP-20 macroporous resin column chromatography,silica gel column chromatography,ODS column chromatography,Sephadex LH-20 column chromatography and high performance liquid chromatography(HPLC).The structures of the compounds were identified according to their spectroscopic data.RESULTS Fifteen phenylpropanoid constituents were isolated from the extract of the stems of Dendrobium officinale and identified as alangilignoside B(1),(-)-(7S,8R)-4,9'-di-O-β-D-glucopyranosyloxydihydrodiconiferyl alcohol(2),5,5'-dimethoxylariciresinol-4-O-β-D-glucoside(3),icariol A2-4-O-β-D-glucoside(4),picraquassioside C(5),syringylgcerol-8-O-4'-(synapyl alcohol)ether(6),(+)-lyoniresionl-9-O-β-D-glucopyranoside(7),liriodendrin(8),syringaresinol-4'-O-β-D-glucoside(9),N-trans-coumaroyl tyramine(10),shashenoside(11),eugenyl-O-β-D-apiofuranosyl-(1″→2')-O-β-D-glucoside(12),3-(4-hydroxy-3,5-dimethoxyphenyl)-propane-1,2-diol(13),dihydroferulic acid(14)and coniferin(15).Compounds 4 and 7 showed weak ABTS radical scavenging capability,and compounds 4,7 and 12 displayed moderate DPPH radical scavenging capability.CONCLUSION Compounds 1,2,7,11-14 are reported from the Dendrobium genus for the first time,and compounds 3,5,6 and 8 have not been previously reported from Dendrobium officinale.Compounds 4,7 and 12 exhibit antioxidant activity.
