摘 要:目的合成金属有机框架ZIF-8-Ce模拟酶,构建一种简单高效比色法来检测谷胱甘肽,并将其运用于人血清中谷胱甘肽(GSH)的检测。方法采用分步合成法,按一定比例掺杂稀土铈,采用透射电子显微镜(transmission electron microscope,TEM)、傅里叶变换红外光谱(fourier transform infrared spectrum,T-IR)、X射线光电子能谱(x-ray photoelectron spectroscopy,XPS)对其结构进行表征。结果所制得的ZIF-8-Ce具有正六面体形貌,表面存在C、N、O、Zn、Ce元素。ZIF-8-Ce具有类似氧化酶的活性,不需要过氧化氢的存在,能催化无色的过氧化物酶底物(TMB)氧化生成蓝色ox-TMB,而GSH能抑制ZIF-8-Ce的氧化酶活性。在最佳条件下,该方法的信号响应在GSH浓度(0.1~25μmol·L^(-1))呈良好的线性关系,r^(2)=0.9928,检测限为23.1 nmol·L^(-1)(S=3δ),并运用于人血清中GSH的检测。结论成功制备ZIF-8-Ce模拟酶,其检测血清GSH的准确度高,重现性好,有潜力成为合适的氧化酶替代物。OBJECTIVE To synthesize metal-organic framework ZIF-8-Ce mimic enzyme,construct a simple and efficient colorimetric method for the detection of glutathion,and apply it to the detection of GSH in human serum.METHODS Rare earth cerium was doped in a certain proportion by step-by-step synthesis,and its structure was characterized by transmission electron microscope(TEM),Fourier transform infrared spectroscopy(FT-IR),and X-ray photoelectron spectroscopy(XPS).RESULTS The prepared ZIF-8-Ce had hexahedron morphology and there were C,N,O,Zn and Ce elements on the surface.ZIF-8-Ce can catalyze the oxidation of colorless peroxidase substrate(TMB)to blue ox-TMB without the presence of hydrogen peroxide,while glutathione(GSH)can inhibit the oxidase activity of ZIF-8-Ce.Under the optimum conditions,the signal response of this method had a good linear relationship at the concentration of GSH(0.1-25μmol·L^(-1)),r^(2)=0.9928,the detection limit was 23.1 nmol·L^(-1)(S=3δ),and the method was applied to the detection of GSH in human serum.CONCLUSION ZIF-8-Ce mimic enzyme is successfully prepared,which has high accuracy and good reproducibility in the detection of serum GSH and has the potential to become a suitable substitute for oxidase.
