摘　　要：目的:探究蛋白磷酸酶1催化亚基α(PPP1CA)蛋白表达与胃癌细胞的增殖及迁移的相关性。方法:选取2016年6月—2021年12月淳安县第一人民医院收治的80例胃癌患者,均采集了胃癌组织和癌旁组织样本,分别命名为胃癌组和癌旁组。比较两组P53、Ki67表达情况,PPP1CA水平,miR-125b表达情况,分析miR-125b、PPP1CA、P53、Ki-67蛋白表达的相关性。结果:胃癌组P53、Ki-67表达阳性率高于癌旁组,差异有统计学意义(P<0.001);胃癌组P53、Ki-67蛋白表达和染色程度高于癌旁组,差异有统计学意义(P<0.05)。免疫组化、WB、PCR 3种检测方式下,胃癌组PPP1CA水平均高于癌旁组,差异有统计学意义(P<0.05)。胃癌组miR-125b表达低于癌旁组,差异有统计学意义(P<0.05)。PPP1CA与P53、Ki-67呈正相关(P<0.05),miR-125b与PPP1CA、P53、Ki-67呈负相关(P<0.05)。结论:miR-125b调控的PPP1CA蛋白表达与胃癌细胞增殖及迁移呈正相关。Objective:To investigate the relationship between the expression of protein phosphatase 1 catalytic subunitα(PPP1CA)protein and the proliferation and migration of gastric cancer cells.Methods:A total of 80 patients with gastric cancer admitted to First People's Hospital of Chun'an County between June 2016 and December 2021 were chosen.Gastric cancer tissue and paracancerous tissue specimens were collected from all patients,which were divided into gastric cancer group and paracancerous group.The expressions of P53 and Ki-67,level of PPP1CA,and expression of miR-125b in the two groups were compared.The relationship between the expressions of miR-125b,PPP1CA,P53,Ki-67 was analyzed.Results:The positive expression rates of P53 and Ki-67 in gastric cancer group were higher than those in paracancerous group,and the difference was statistically significant(P<0.001);The expression of P53 and Ki-67 proteins and staining degree in the gastric cancer group were higher than those in the paracancerous group,and the difference was statistically significant(P<0.05).By the three detection methods of immunohistochemistry,WB and PCR,the PPP1CA level in the gastric cancer group was higher than that in the paracancerous group,and the difference was statistically significant(P<0.05).The expression of miR-125b in the gastric cancer group was lower than that in the paracancerous group,and the difference was statistically significant(P<0.05).PPP1CA was positively correlated with P53 and Ki-67(P<0.05),and miR-125b was negatively correlated with PPP1CA,P53 and Ki-67(P<0.05).Conclusion:The expression of PPP1CA protein regulated by miR-125b is positively correlated with the proliferation and migration of gastric cancer cells.
