摘 要:为了降低γ-聚谷氨酸(γ-PGA)的生产成本,实现芦苇等可再生资源的高值化利用,探索建立以芦苇水解液为碳源制备γ-PGA的新型发酵过程控制策略的可行性。首先,研究纤维素酶用量和底物浓度对芦苇水解的影响;其次,采用分段式培养及溶氧反馈补料调控策略,对芦苇水解液发酵生产γ-PGA进行工艺优化。结果表明,芦苇经预处理后在10 FPU/g(底物)酶用量和10%底物添加量的条件下水解,总糖质量浓度可达(76.64±1.74)g/L;以分段式溶氧反馈-分批补料发酵培养方式,利用芦苇水解液生产γ-PGA,最终产量可达(46.72±1.18)g/L,生产效率较分批发酵提高了46.06%。因此,将芦苇水解液用于γ-PGA的分批补料发酵具有产业化可行性,有助于降低商业化生产的原料成本。In order to reduce the production cost ofγ-polyglutamic acid(γ-PGA)and realize high-value utilization of renewable resources such as reed,the feasibility of establishing a new fermentation process control strategy for the preparation ofγ-PGA using reed hydrolysate as carbon source was explored.Firstly,the effects of cellulase dosage and substrate concentration on reed hydrolysis were studied;Secondly,the process of fermentation and production ofγ-PGA from reed hydrolysate was optimized by segmented culture and DO feed-back control strategies.The results show that after pretreatment,the total sugar concentration of reed could reach(76.64±1.74)g/L under the condition of 10 FPU/g(substrate)enzyme dosage and 10%substrate addition;the reed hydrolysate was producedγ-PGA by segmented dissolved oxygen feedback-batch feed fermentation culture,and the final yield could reach(46.72±1.18)g/L,with the production efficiency increasing by 46.06%compared with batch fermentation.The use of reed hydrolysate for batch fermentation ofγ-PGA is industrially feasible,which helps to reduce the raw material cost of its commercial production.
