摘  要:目的:探究调控微小RNA(miR)-642a-5p表达对宫颈癌HeLa细胞增殖、侵袭和凋亡的影响及机制。方法:通过转染不同质粒将Hela细胞分为miR-642a-5p mimic组、miR-642a-5p mimic NC组,以未做任何处理的Hela细胞作为对照组。RT-qPCR法检测miR-642a-5p、NF-κB mRNA的表达水平,Western blot法检测NF-κB p65蛋白的表达水平,MTT法检测细胞增殖活力,Transwell法检测细胞侵袭、迁移能力,流式细胞术检测细胞凋亡情况。结果:与对照组、miR-642a-5p mimic NC组比较,miR-642a-5p mimic组miR-642a-5p的表达量及细胞凋亡率升高,NF-κB mRNA和NF-κB p65蛋白的表达水平及转染24、48 h的细胞增殖活性、侵袭细胞个数、划痕愈合率降低(均P<0.05);对照组与miR-642a-5p mimic NC组miR-642a-5p、NF-κB mRNA、NF-κB p65蛋白的表达水平及转染24、48 h细胞增殖活性、侵袭细胞个数、划痕愈合率、细胞凋亡率比较,差异无统计学意义(均P>0.05)。对照组、miR-642a-5p mimic NC组细胞增殖活性呈时间依赖性升高,miR-642a-5p mimic组细胞增殖活性呈时间依赖性下降(均P<0.05)。结论:高表达的miR-642a-5p可抑制宫颈癌细胞的增殖、侵袭和迁移能力,促进细胞凋亡,这一机制可能与抑制NF-κB p65蛋白表达有关。Objective:To investigate the effects of microRNA(miR)-642a-5p expression on the proliferation,invasion and apoptosis of cervical cancer HeLa cells and its mechanism.Methods:The Hela cells were divided into miR-642a-5p mimic group,miR-642a-5p mimic NC group by transfection with different plasmids,and the Hela cells without any treatment were used as control group.The expression of miR-642a-5p and NF-κB mRNA in the three groups of cells was detected by qRT-PCR,the expression level of NF-κB p65 protein in the three groups of cells was detected by Western blot,the proliferative vigour of the three groups was detected by MTT,the invasive and migratory capacity of the three groups of cells was detected by Transwell,and the apoptosis of the three groups was detected by flow cytometry.Results:Compared with control group and miR-642a-5p mimic NC group,miR-642a-5p mimic group showed higher expression of miR-642 a-5p and apoptosis rate,and lower expression of NF-κB mRNA and NF-κB p65 protein as well as proliferative activity of the cells transfected for 24,48 hours,the number of invasive cells,and the healing rate of the scratch of transfection(all P<0.05).The expressions of miR-642a-5p,NF-κB mRNA,NF-κB p65 protein and the proliferative activity of cells transfected for 24 and 48 hours,the number of invading cells,the rate of scratch healing,and the rate of apoptosis in the control group in comparison with those in the miR-642a-5p mimic NC group,and the difference was not statistically significant(all P>0.05).There was a time-dependent increase in cell proliferation activity in the control and miR-642a-5p mimic NC groups,and a time-dependent decrease in cell proliferation activity in the miR-642a-5p mimic group(all P<0.05).Conclusion:High expression of miR-642a-5p inhibits the proliferation,invasion and migration ability of cervical cancer cells and promotes apoptosis,a mechanism that may be related to the inhibition of the expression of NF-κB p65 protein.
