摘 要:目的:探究肿瘤坏死因子受体相关因子6(TRAF6)高表达对子宫内膜癌细胞侵袭、迁移及上皮间质转化的影响。方法:将Ishikawa细胞随机分为三组:对照组(子宫内膜癌细胞Ishikawa未转染)、NC组(子宫内膜癌细胞Ishikawa转染空载质粒)和TRAF6高表达组(子宫内膜癌细胞Ishikawa转染TRAF6过表达质粒);实时荧光定量PCR(RT-qPCR)和蛋白免疫印迹(WB)检测TRAF6 mRNA以及蛋白表达情况;Transwell实验、划痕实验以及裸鼠移植瘤实验检测细胞侵袭、迁移能力;WB检测上皮间质转化标志蛋白E型钙黏蛋白(E-cadherin)、闭合蛋白(Occludin)、N型钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)表达水平。结果:相较于人子宫内膜上皮细胞,子宫内膜癌细胞Ishikawa中TRAF6 mRNA和蛋白表达显著降低(均P<0.05)。相较于对照组和NC组,TRAF6高表达组的TRAF6 mRNA水平和蛋白表达显著升高,细胞侵袭、迁移能力和Vimentin、N-cadherin蛋白表达显著降低,Occludin、E-cadherin蛋白表达显著升高(均P<0.05);对照组和NC组上述指标比较差异无统计学意义(均P>0.05)。结论:上调子宫内膜癌细胞中TRAF6的表达,可抑制上皮间质转化过程,减弱子宫内膜癌细胞的侵袭和迁移能力,TRAF6在子宫内膜的发生发展中具有重要的调节作用。Objective:To explore the effects of high expression of tumor necrosis factor receptor-associated factor 6(TRAF6)on the invasion,migration and epithelial-mesenchymal transition of endometrial cancer cells.Methods:Ishikawa cancer cells were randomly divided into three groups:control group(endometrial cancer Ishikawa cells were not transfected),NC group(endometrial cancer Ishikawa cells were transfected with empty plasmid)and TRAF6 high expression group(endometrial cancer Ishikawa cells were transfected with TRAF6 overexpression plasmid).Real-time fluorescent quantitative PCR(RT-qPCR)and Western blot(WB)were used to detect TRAF6 mRNA and protein expression.Transwell experiment,scratch experiment and nude mouse xenograft experiment were used to detect cell invasion and migration abilities.WB was used to detect the expression levels of markers of epithelial-mesenchymal transition,including E-cadherin,Occludin,N-cadherin and Vimentin.Results:Compared with human endometrial epithelial cells,the expression of TRAF6 mRNA and protein in endometrial cancer cells Ishikawa were significantly decreased(all P<0.05).Compared with the control group and NC group,the TRAF6 mRNA level and protein expression in the TRAF6 high expression group were significantly increased,cell invasion and migration abilities,Vimentin and N-cadherin protein expressions were significantly reduced,and Occludin and E-cadherin protein expressions were significantly increased(all P<0.05).There was no statistically significant difference in the above indicators between the control group and the NC group(all P>0.05).Conclusion:Up-regulating the expression of TRAF6 in endometrial cancer cells can inhibit the epithelial-mesenchymal transition process and weaken the invasion and migration abilities of endometrial cancer cells.
