摘 要:目的 观察决明子提取物调控骨骼肌LKB1-AMPK-GLUT4信号通路改善糖尿病大鼠胰岛素抵抗的效果。方法 采用高脂饮食联合链脲佐菌素(STZ)建立2型糖尿病大鼠模型,选择健康大鼠10只为正常组,40只建模成功大鼠分为模型组、高剂量决明子提取物组(高剂量组)、中剂量决明子提取物组(中剂量组)和低剂量决明子提取物组(低剂量组)各10只,高剂量组给予450 mg/(kg·d)、中剂量组给予150 mg/(kg·d)和低剂量组给予50 mg/(kg·d)决明子提取物灌胃,连续治疗12周,正常组及模型组均给予等体积生理盐水灌胃。12周后采用实时荧光定量PCR、蛋白免疫印迹法检测大鼠腓肠肌LKB1-AMPK-GLUT4信号通路的肝脏激酶B1 (LKB1)、腺苷酸活化蛋白激酶(AMPK)和葡萄糖转运蛋白-4 (GLUT4)基因及蛋白的相对表达量,采用高胰岛素-正糖钳夹试验(HEC)、胰岛素耐量试验(IPITT)综合评价胰岛素抵抗水平。采用酶联接免疫吸附剂测定法(ELISA)检测大鼠腓肠肌(骨骼肌)组织中氧化应激指标。结果 与正常组比较,模型组大鼠体质量明显降低,空腹血糖及空腹胰岛素(FINS)水平明显增高,差异均有统计学意义(P<0.05);与模型组比较,决明子各给药组体质量明显增加,空腹血糖及FINS水平明显降低,差异均有统计学意义(P<0.05),且各给药组间比较呈剂量依赖性。与正常组比较,模型组大鼠葡萄糖输注率(GIR)水平明显降低,差异具有统计学意义(P<0.05);与模型组比较,决明子各给药组GIR水平明显增加,差异具有统计学意义(P<0.05),且各给药组间比较呈剂量依赖性。与正常组比较,模型组大鼠各时点的胰岛素耐量均明显增加,差异具有统计学意义(P<0.05);与模型组比较,决明子各给药组大鼠各时点的胰岛素耐量均明显降低,差异具有统计学意义(P<0.05),且各给药组间比较呈剂量依赖性。与正常组比较,模型组大鼠谷胱甘肽(GSH)及超氧化物歧化酶(SOD)水平明�Objective To observe the effect of cassia seed extract on improving insulin resistance in diabetes rats by regulating LKB1-AMPK-GLUT4 signaling pathway in skeletal muscle.Methods The rat model of type 2 diabe-tes was established by high-fat diet combined with streptozotocin(STZ).Ten healthy rats were selected as the normal group,and 40 successfully modeled rats were divided into model group,high-dose cassia seed extract group,medi-um-dose cassia seed extract group,and low-dose cassia seed extract group(with 10 rats in each group),which were giv-en 450 mg/(kg·d),150 mg/(kg·d),and 50 mg/(kg·d)cassia seed extract by gavage for 12 weeks of continuous treat-ment,respectively.Both normal group and model group were given an equal volume of normal saline.After 12 weeks,real-time quantitative PCR and Western blotting were used to detect the relative expression of LKB1-AMPK-GLUT4 sig-naling pathway-related genes and proteins in rat gastrocnemius muscle(bone muscle).High insulin-negative glucose clamp test and insulin tolerance test were used to comprehensively evaluate insulin resistance levels.The changes of oxi-dative stress indicators in rat skeletal muscle tissues were detected by enzyme-linked immunosorbnent assay(ELISA).Results Compared to normal group,levels of body mass of model group rats were significantly reduced,and its levels of fasting blood glucose and fasting insulin(FINS)were significantly increased,with statistically significant differences(P<0.05);compared to model group,levels of body mass of high-dose,medium-dose,low-dose cassia seed extract groups were significantly increased,and their fasting blood glucose and FINS levels were significantly decreased,with statistically significant differences(P<0.05),in a dose-dependent manner.Compared to normal group,levels of glucose infusion rate(GIR)of model group were significantly reduced(P<0.05);compared to model group,levels of GIR of high-dose,medium-dose,low-dose cassia seed extract groups were significantly increased(P<0.05),in a dose-dependent manner.C
