柿叶总黄酮抑制巨噬细胞源性泡沫细胞脂质蓄积的作用研究

摘　　要：目的探讨柿叶总黄酮对巨噬细胞源性泡沫细胞胆固醇脂质蓄积的影响及其潜在机制。方法体外培养THP-1细胞,160nmol/L佛波酯诱导分化为巨噬细胞,与50μg/mL ox-LDL孵育建立巨噬细胞源性泡沫细胞模型,分别用不同浓度的柿叶总黄酮(0、0.5、1、2、4μg/mL)处理。油红O染色检测细胞内脂质蓄积情况,高效液相色谱法检测细胞内胆固醇含量,qPCR和Western blot分别检测SR-A和ABCA1 mRNA和蛋白表达情况。结果柿叶总黄酮能显著性抑制巨噬细胞源性泡沫细胞脂质蓄积,减少细胞内游离胆固醇(FC)、总胆固醇(TC)和胆固醇酯(CE)含量,降低相关蛋白SR-A的摄取以及促进胆固醇外排关键蛋白ABCA1表达。结论柿叶总黄酮通过降低相关蛋白SR-A的摄取以及促进胆固醇外排关键蛋白ABCA1表达,减少细胞内胆固醇含量,抑制巨噬细胞源性泡沫细胞脂质蓄积。Objective To explore the effect and underlining mechanisms of flavonoids from persimmon leaf(FPL)on lipid accumulation in macrophage-derived foam cells.Methods THP-1 cells were differentiated into macrophages by 160 nmol/L phorbol myristate acetate and incubated with 50μg/L oxidized low density lipoprotein to induce foam cells to establish macrophage-derived foam cells model.The intracellular lipid accumulation was detected by Oil red O staining.The cholesterol contents were tested by High Performance Liquid Chromatogram method.The quantitative PCR and Western Blot were used to measure the mRNA and protein expression of SR-A and ABCA1.Results FPL obviously inhibited the intracellular lipids accumulation,reduced the level of free cholesterol(FC),total cholesterol(TC)and cholesterol ester(CE),suppressed the SR-A expression,and increased the ABCA1 expression and cholesterol efflux in THP-1 macrophages-derived foam cells.Conclusion FPL reduces the cholesterol contents through suppression the SR-A expression,the relative cholesterol uptake protein,and increasing the ABCA1 expression that a important protein involved in cholesterol efflux,to inhibit lipid accumulation in THP-1 macrophages-derived foam cells.
