摘  要:目的 探讨沉默原癌基因人类免疫缺陷病毒短转录诱导物连接因子1(FBI-1)对肺腺癌A549细胞增殖、迁移和侵袭的影响及机制。方法 qRT-PCR法检测人正常肺上皮细胞株CCD-8L和人非小细胞肺癌细胞株A549中FBI-1 mRNA表达水平。将A549细胞分为对照组、shRNA-NC组、shRNA-FBI-1组、shRNA-FBI-1+TGF-β1组,各组细胞分别转染对应的慢病毒,shRNA-FBI-1+TGF-β1组细胞转染慢病毒后,10 ng/mL转化生长因子β1(TGF-β1)处理24 h,MTT法检测细胞存活率、Transwell实验检测细胞侵袭和迁移能力,蛋白印迹法检测FBI-1、TGF-β1、p-Smad2、上皮钙粘蛋白(E-cad)、神经钙粘蛋白(N-cad)和波形蛋白(Vimentin)相对表达水平。结果 肺癌细胞A549中FBI-1 mRNA表达升高;与对照组和shRNA-NC组比较,shRNA-FBI-1组细胞存活率、迁移和侵袭能力以及FBI-1、TGF-β1、p-Smad2、N-cad和Vimentin蛋白表达降低,E-cad蛋白表达升高(P<0.05);与shRNA-FBI-1组比较,shRNA-FBI-1+TGF-β1组细胞存活率、迁移和侵袭能力、FBI-1、TGF-β1、p-Smad2、N-cad和Vimentin蛋白表达升高,E-cad蛋白表达降低(P<0.05)。结论 沉默FBI-1可抑制肺癌A549细胞增殖、迁移和侵袭,其可能是通过抑制TGF-β1/Smads信号通路发挥作用。Objective To investigate the effects of human immunodeficiency virus short transcription inducer ligand 1(FBI-1)on proliferation,migration and invasion of lung adenocarcinoma A549 cells and its mechanism.Methods qRT-PCR was used to detect FBI-1 mRNA expression in human normal lung epithelial cell line CCD-8L and human non-small cell lung cancer cell line A549.A549 cells were divided into control group,shRNA-NC group,shRNA-FBI-1 group and shRNA-FBI-1+TGF-β1 group.Cells in each group were transfected with the corresponding lentivirus,and cells in shRNA-FBI-1+TGF-β1 group were transfected with lentivirus.After 10 ng/mL transforming growth factorβ1(TGF-β1)treatment for 24 h,the cell survival rate was detected by MTT assay,and the invasion and migration of cells were detected by Transwell assay.Western blot was used to detect the relative expression levels of FBI-1,TGF-β1,p-Smad2,epithelial cadherin(E-cad),neuro cadherin(N-cad)and Vimentin.Results The expression of FBI-1 mRNA was increased in lung cancer cells A549.Compared with the control group and shRNA-NC group,the cell survival rate,migration and invasion ability,and the protein expression of FBI-1,TGF-β1,p-Smad2,N-cad and Vimentin were decreased in shRNA-FBI-1 group,while the protein expression of E-cad was increased(P<0.05).Compared with shRNA-FBI-1 group,the expression of cell survival rate,migration and invasion ability,FBI-1,TGF-β1,p-Smad2,N-cad and Vimentin protein was increased in shRNA-FBI-1+TGF-β1 group,while the expression of E-cad protein was decreased(P<0.05).Conclusion FBI-1 silencing can inhibit the proliferation,migration and invasion of lung cancer A549 cells,possibly by inhibiting the TGF-β1/Smads signaling pathway.
