IMP-3和Cyclin B1在鼻咽癌中的表达水平及临床意义

摘　　要：目的 探讨胰岛素样生长因子ⅡmRNA结合蛋白3(IMP-3)和细胞周期蛋白B(Cyclin B1)在鼻咽癌(NPC)中的表达水平及临床意义。方法 回顾性选取NPC患者56例,另选取同期行鼻咽部活检病理证实为慢性鼻咽炎(CN)的组织样本26例作为对照组。采用免疫组化法检测2组中IMP-3和Cyclin B1的表达,分别分析蛋白表达与NPC患者临床病理特征的关系,采用Spearman等级相关分析法分析IMP-3和Cyclin B1蛋白表达的相关性。结果 IMP-3蛋白在NPC组织中的阳性表达率为75.00%,在CN组织中的阳性表达率为1.61%;Cyclin B1蛋白在NPC组织中的阳性表达率为69.64%,在CN组织中的阳性表达率为7.69%;2组间IMP-3和Cyclin B1蛋白的阳性表达率比较,NPC组均高于CN组,差异有统计学意义(P<0.05)。NPC组织中IMP-3蛋白的高表达与年龄、TNM分期有相关性(P<0.05),Cyclin B1蛋白的高表达与TNM分期、淋巴结转移有相关性(P<0.05);经Spearman等级相关分析法分析,IMP-3和Cyclin B1蛋白表达无相关性(γ=0.195,P=0.102)。结论 NPC组织中IMP-3和Cyclin B1蛋白均呈现高表达,IMP-3和Cyclin B1蛋白与NPC的发生及发展密切相关。Objective To investigate the expression and clinical significance of insulin-like growth factorⅡmRNA binding protein 3(IMP-3)and cyclin B1 in nasopharyngeal carcinoma(NPC).Methods 56 cases of NPC patients were retrospectively selected,and 26 cases of tissue samples confirmed as chronic nasopharyngitis(CN)by nasopharyngeal biopsy at the same time were selected as the control group.Immunohistochemical method was used to detect the expression of IMP-3 and CyclinB1 in the 2 groups,and the relationship between protein expression and clinicopathological characteristics of NPC patients was analyzed.Spearman rank correlation analysis was used to analyze the correlation between IMP-3 and CyclinB1 protein expression.Results The positive expression rate of IMP-3 protein was 75.00%in NPC and 1.61%in CN.The positive rates of IMP-3 and CyclinB1 in NPC and CN were 69.64%and 7.69%respectively.The positive rates of IMP-3 and CyclinB1 in NPC were higher than those in CN(P<0.05).The high expression of IMP-3 protein was correlated with age and TNM stage(P<0.05).The high expression of CyclinB1 protein was correlated with TNM stage and lymph node metastasis(P<0.05).There was no correlation between IMP-3 and CyclinB1 protein expression by Spearman rank correlation analysis(γ=0.195,P=0.102).Conclusion IMP-3 and CyclinB1 protein are highly expressed in NPC tissues,which are closely related to the occurrence and development of NPC,and may become the diagnostic and prognostic markers of NPC patients.
