摘 要:目的:探讨MMI-166对人胰腺癌细胞PANC-1和SW1990迁移和侵袭的影响。方法:培养人胰腺癌细胞PANC-1、SW1990,使用不同浓度MMI-166进行干预,细胞划痕实验和Transwell实验检测细胞迁移和侵袭能力。比较各组PANC-1及SW1990细胞中MMP-2及MMP-9蛋白表达量。结果:与对照组相比,通过细胞划痕实验和Transwell实验,发现MMI-166对胰腺癌细胞系的细胞迁移能力和侵袭能力,随着浓度增加而下降明显,差异有统计学意义(P<0.05);Western blot及免疫荧光显示,MMI-166对胰腺癌细胞系MMP-2及MMP-9蛋白的表达成显著抑制作用,且存在剂量依赖性,差异有统计学意义(P<0.05)。结论:MMI-166通过特异性抑制MMP-2和MMP-9表达,在胰腺癌细胞的迁移和侵袭调控中发挥重要作用。Objective:To investigate the effects of MMI-166 on the migration and invasion of human pancreatic cancer cell lines PANC-1 and SW1990.Methods:PANC-1,SW1990 cells were cultured in vitro,which were treatment with different concentrations of MMI-166.Cell migration and invasion were detected by scratch assay and Transwell assay.The expressions of proteins of MMP-2 and MMP-9 were compared in different groups of PANC-1 and SW1990 cells.Results:Compared with the control group,the scratch assay and Transwell assay showed that MMI-166 decreased cell migration and invasion with increasing concentration,with statistical difference(P<0.05).Western blot and immunofluorescence showed MMI-166 significantly inhibited the expression of MMP-2 and MMP-9 protein in a dose-dependent manner,with statistical difference(P<0.05).Conclusion:MMI-166 inhibited the MMP-2 and MMP-9 activity in pancreatic cancer cells and it is useful for the regulation of pancreatic cancer cells migration and invasion.
