摘  要:目的探讨皮肤鳞状细胞癌(CSCC)进展的生物学标志物。方法在GEO数据库中筛选CSCC、日光性角化(AK)和正常皮肤组织的基因表达谱芯片数据集,通过GEO2R在线数据库筛选在正常组织、AK和CSCC3组间存在显著表达差异的基因。纳入2022年6月—12月本院皮肤科65例CSCC患者(CSCC组)、40例AK患者(AK组)和30例正常对照者(对照组)作为研究对象,组织和血清中目的蛋白的表达水平采用免疫组化法和ELISA法检测。血清中目的蛋白表达水平在鉴别AK和CSCC的临床效能采用受试者操作特征曲线(ROC)分析。结果GEO2R在线数据库筛选出5个共同差异表达基因TMSΒ10、PDZD2、ENO1、RRBP1和S100A11。ENO1和S100A11蛋白表达水平在对照组、AK组和CSCC组3组间两两相比差异有统计学意义(P<0.05)。CSCC患者血清中ENO1和S100A11蛋白表达水平与肿瘤细胞分化程度和浸润深度显著相关(P<0.05)。ROC曲线后显示,ENO1和S100A11蛋白表达水平在鉴别AK和CSCC的AUC分别为0.964和0.953,其中ENO1的截断值为35.12 ng/ml,诊断敏感度和特异度分别为95.13%和92.45%,其中S100A11的截断值为23.68 ng/ml,诊断敏感度和特异度分别为94.67%和93.21%。免疫组化结果显示,ENO1在癌组织高度表达18例,中度表达12例,癌旁组织中均无阳性表达;S100A11在癌组织高度表达30例,癌旁组织中均无阳性表达。与癌旁组织相比,CSCC癌组织中ENO1和S100A11阳性表达率显著升高(P<0.05)。结论ENO1和S100A11可能是促进CSCC进展的生物学标志物。Objective This paper aims to investigate the progression biomarkers of cutaneous squamous cell carcinoma(CSCC).Methods The microarray data sets of gene expression profiles of CSCC,Actinic Keratosis(AK)and normal skin tissue were screened in GEO database.The significant differently expressed genes among normal tissue,AK and CSCC were screened by GEO2R online database.A total of 65 patients with CSCC(CSCC group),40 patients with AK(AK group)and 30 normal controls(control group)were enrolled between June 2022 and December 2022 in the Department of Dermatology in our hospital.The levels of target proteins in tissues and serum were detected by immunohistochemical and ELISA methods.The clinical efficacy of serum target protein expression levels in distinguishing AK from CSCC was analyzed by receiver operating characteristic curve(ROC).Results Five common differentially expressed genes including TMSB10,PDZD2,ENO1,RRBP1 and S100A11 were identified in GEO2R online database.The differences in the serum levels of ENO1 and S100A11 were statistically significant among control group,AK group and CSCC group(P<0.05).The serum expression levels of ENO1 and S100A11 in CSCC patients were significantly correlated with the degree of differentiation and the depth of invasion of tumor cells(P<0.05).ROC curve showed that the AUC of serum ENO1 and S100A11 levels were 0.964 and 0.953 for distinguis⁃hing AK from CSCC.The cut-off value of ENO1 was 35.12 ng/ml,and sensitivity and specificity were 95.13%and 92.45%,respectively.The cut-off value of S100A11 was 23.68 ng/ml,and sensitivity and specificity were 94.67%and 93.21%,respectively.Immunohistochemical results showed that ENO1 were highly expressed in tumor tissues in 18 cases,moderately expressed in tumor tissues in 12 cases,and no positive expression was found in paracancer tissues.S100A11 was highly expressed in 30 cases of tumor tissue,but no positive expression was found in paracancer tissue.The positive expression rates of ENO1 and S100A11 in tumor tissues were significantly highe
