摘  要:目的观察大蒜素对高糖环境人心脏成纤维细胞(HCFs)增殖和凋亡的影响及其作用机制。方法贴壁法培养HCFs,体外建立高糖诱导的HCFs增殖模型,分为对照组(5.5 mmol/L)、高糖组(25 mmol/L)以及高糖联合不同浓度的大蒜素组(分别为2.5μg/mL、5μg/mL、10μg/mL、20μg/mL)。应用MTT比色、瑞氏-吉姆萨染色倒置生物显微镜、Hochest 33258染色荧光显微镜、流式细胞分析技术、半胱氨酸蛋白酶-3(Caspase-3)活性测定、Western blot检测等方法观察大蒜素对HCFs增殖和凋亡的作用。结果与对照组比较,高糖组细胞增殖增加,B细胞淋巴瘤-2(Bcl-2)增加,Bcl-2相关X蛋白(Bax)降低。与高糖组比较,大蒜素各组细胞增殖降低,细胞凋亡率及Casepas-3水平提高,高糖联合10μg/mL大蒜素组及高糖联合20μg/mL大蒜素组Bcl-2减低,Bax增加,差异均有统计学意义(P<0.05)。不同浓度大蒜素处理高糖环境HCFs 72 h,可见到细胞增殖受到抑制,细胞数较高糖组有所减少,染色质浓缩、边集、碎裂,凋亡细胞增多。结论大蒜素抑制高糖环境HCFs的增殖,促进其凋亡,其机制可能与下调Bcl-2、上调Bax、增强Caspase-3蛋白活性有关。Objective To explore the effect of allicin on the proliferation and apoptosis of human cardiac fibroblasts(HCFs)in high glucose environment and its mechanism.Methods HCFs were cultured using an adherent method.A model of HCFs proliferation under high glucose induction was established in vitro,with a control group(5.5 mmol/L),a high glucose group(25 mmol/L),and high glucose groups treated with different concentrations of allicin(2.5μg/mL,5μg/mL,10μg/mL,and 20μg/mL).The effects of allicin on the proliferation and apoptosis of HCFs were assessed using MTT colorimetry,inverted biomicroscopy with Riesling Giemsa staining,fluorescence microscopy with Hochester 33258 staining,flow cytometry,Caspase-3 activity assay,and Western blot.Results Compared with the control group,the high glucose group showed increased cell proliferation,upregulation of B-cell lymphoma-2(Bcl-2),and downregulation of Bcl-2 associated X protein(Bax).Compared with the high glucose group,allicin-treated groups exhibited decreased cell proliferation,elevated apoptosis rate and Caspase-3 level,and the high-glucose groups treated with 10μg/mL and 20μg/mL allicin showed decreased Bcl-2 and increased Bax levels,with statistically significant difference(P<0.05).After 72 h treatment with different concentrations of allicin in a high glucose enviroment,cell proliferation was inhibited,cell number was reduced compared to the high-glucose group;chromatin condensation,margination,fragmentation,and an increase in apoptotic cells were observed.Conclusion Allicin inhibits the proliferation of HCFs in high glucose environments,and promotes apoptosis.The mechanism may be related to the downregulation of Bcl-2,upregulation of Bax,and enhanced Caspase-3 protein activity.
