摘 要:目的探索卵巢癌组织中差异性miRNAs并通过基础实验探索其分子机制。方法使用R语言获取GEO数据库中GSE61485(癌旁组织5例,癌组织5例),GSE71477(癌旁组织4例,癌组织4例)和GSE106817(癌旁组织65例,癌组织325例)表达谱数据,癌旁作为对照组,做差异分析,Starbase工具做生存分析。选取卵巢癌SKOV3细胞系,构建miR-150-5p过表达、敲低和阴性对照细胞模型,并用CCK8实验对增殖能力评估,通过LinkedOmics做miR-150-5p的相关性分析以及富集分析。结果分别对GSE61485,GSE71477和GSE106817表达谱数据差异分析和卵巢癌的预后分析发现,miR-150-5p是唯一共有的肿瘤组织下调基因;并且miR-150-5p过表达SKOV3细胞的增殖能力明显下降(P<0.01),而抑制miR-150-5p表达后,SKOV3细胞增殖能力增强(P<0.01);进一步通过LinkedOmics工具对miR-150-5p相关性基因分析发现负相关基因中PTCH1相关性最强(P<0.01,R=-0.47),正相关基因中IL-2RG相关性最强(P<0.01,R=0.76);此外GO、KEGG富集分析发现miR-150-5p与免疫过程、细胞黏附以及代谢过程密切相关。结论miR-150-5p作为卵巢癌发生发展过程中的一个关键分子参与卵巢癌细胞生长等过程,有望成为一个新的治疗靶点。Objective To explore the differential miRNAs in ovarian cancer tissues and explore their molecular mechanism and target feasibility through basic experiments.Methods GSE61485,GSE106817 and GSE71477 data sets were acquired by R language.The paracancer tissue served as the control group.Survival analysis was performed by Starbase.Ovarian cancer SKOV3 cell line was selected to construct miR-150-5p overexpression,knockdown and negative control cell models,and CCK8 assay was used to evaluate the proliferation ability.Correlation analysis and enrichment analysis of miR-150-5p were performed on LinkedOmics.Results Differential analysis of GSE61485,GSE71477 and GSE106817 expression profiles and prognostic analysis of ovarian cancer showed that miR-150-5p was the only common down-regulated gene in tumor tissue.Moreover,the proliferation ability of SKOV3 cells with miR-150-5p overexpression was decreased significantly(P<0.01),while the proliferation ability of SKOV3 cells was enhanced after miR-150-5p expression was inhibited(P<0.01).The LinkedOmics tool was used to analyze the miR-150-5p correlation genes.It was found that PTCH1 had the strongest correlation in the negative correlation genes(P<0.01,r=-0.47),and IL-2RG had the strongest correlation in the positive correlation genes(P<0.01,r=0.76).In addition,GO and KEGG enrichment analysis showed that miR-150-5p was closely related to immune processes,cell adhesion and metabolic processes.Conclusion miR-150-5p is a key molecule involved in the growth of ovarian cancer cells which may become a target on clinical treatment.
