摘 要:线粒体是重要的细胞器,参与细胞代谢的调控,决定细胞的命运。而三羧酸循环是产生细胞能量和生物合成的枢纽。多种癌症具有三羧酸循环相关酶的突变,如异柠檬酸脱氢酶、琥珀酸脱氢酶和富马酸水合酶,导致三羧酸循环中间代谢物含量失调,并可能与癌症的发生有关。三羧酸循环中间代谢物作为一种信号分子,通过调节细胞活动参与调控特定转录因子、调节表观遗传、参与染色质修饰,最终通过调控基因表达决定细胞功能,影响肿瘤的发生和发展过程。As important organelles,mitochondria participate in the regulation of cell metabolism and determine the fate of cells.The tricarboxylic acid cycle has been considered as a hub for generating cellular energy and biosynthesis.Multiple cancers have mutations in enzymes involved in tricarboxylic acid cycle,such as isocitrate dehydrogenase,succinate dehydrogenase,and fumarate hydratase,which lead to imbalance of tricarboxylic acid cycle metabolites content and may be associated with cancer.By regulating cell activity,tricarboxylic acid cycle metabolites act as signal molecules,regulating specific transcription factors and epigenetics,and modifying chromatin.Eventually,they determine cell functions by regulating gene expression,and affect the occurrence and development of tumors.
