摘 要:目的 对安徽省蜱类种群分布调查中发现的一新纪录种进行鉴定。方法 对2022年安徽省合肥市蜱类分布调查中采集的蜱标本进行形态学鉴定后,采用PCR方法扩增蜱线粒体16S rRNA基因和细胞色素C氧化酶亚基I(COI)基因,测序后进行同源性分析,用Mega11.0软件构建系统发生树并进行系统进化分析。结果 根据蜱的须肢、假头基、盾板、缘垛、气门板、肛沟、内距及生殖孔等形态特征,鉴定为褐黄血蜱;16S rRNA基因序列与已知褐黄血蜱16S rRNA基因同源性为95.95%~99.51%,与其他蜱种同源性为83.02%~93.47%;COI基因序列与已知的褐黄血蜱COI基因同源性为99.52%~100%,与其他蜱种同源性为82.85%~91.27%。系统发生树表明,待检蜱与已知的褐黄血蜱聚在一起,形成独立的分支。结论 在传统形态学分类鉴定的基础上结合分子生物学鉴定,能更准确地鉴定蜱的种类,也能更好地了解其进化关系,为蜱传疾病的预防控制提供依据。Objective To identify a newly recorded species discovered in the tick investigation in Anhui province,China.Methods After morphological identification of a specimen found during the investigation of tick distribution in Hefei,Anhui province,mitochondrial 16S rRNA gene and cytochrome C oxidase subunit I(COI) gene were amplified by PCR,sequenced for homology analysis.Mega11.0 software was used to construct the phylogenetic trees for phylogenetic analysis.Results According to morphological characteristics,the Ixodidae was morphologically identified as Haemaphysalis flava.The homology of 16SrRNA gene sequence with known 16S rRNA gene of Haemaphysalis flava was 95.95%-99.51%;in comparison with other ticks of Ixodidae,the homology was 83.02%-93.47%.The homology of COI gene sequence with known COI gene of Haemaphysalis flava was 99.52%-100%.The homology was 82.85%-91.27% in comparison with other ticks of Ixodidae.The phylogenetic trees showed that the tick to be identified was clustered together with the known Haemaphysalis flava and formed independent branches.Conclusion It can more accurately identify tick species by combination of traditional morphological classification and molecular biological identification methods,contributing to better understanding of the evolutionary relationships of ticks.Further phylogenetic analysis of gene sequences also provides a basis for the prevention and control of tick-borne diseases.
