摘 要:肿瘤发展是一个多步骤的过程,通过持续与肿瘤微环境(TME)相互作用,构建重要的代谢关系。在TME中,肿瘤细胞主要通过有氧糖酵解完成能量补给,即“Warburg效应”。通过调节代谢关键酶的表达满足异常增殖的肿瘤细胞生物合成的高代谢需求。乳酸脱氢酶A(LDHA)是糖酵解过程中的关键酶之一,通过将丙酮酸转化为乳酸,提高肿瘤细胞糖酵解水平,导致乳酸产量增加,进而促进TME酸化,增强免疫抑制细胞的表达,最终影响TME中的免疫反应。因此,LDHA有望成为增强肿瘤免疫治疗效果的潜在靶点,同时需要进一步开展相关靶向药物的研发。Tumor development is a multi-step process,which continuously interacts with tumor microenvironment(TME)to form a critical metabolic nexus.The phenomenon of aerobic glycolysis is known as the Warburg effect,which contributes to most tumor energy supply,regulating the expression of key metabolic enzymes to fulfill elevated metabolic demands for cellular biosynthesis of abnormally proliferating tumor cells.Lactate dehydrogenase A(LDHA)is one of the key enzymes in glycolysis,which facilitates glycolytic process by converting pyruvate to lactate,improving the glycolysis level of tumor cells,leading to an increased lactate production,acidification of TME,and enhancement of immunosuppressive cell,finally affecting the immune response in the TME.Therefore,LDHA is expected to become a potential target to enhance the efficacy of tumor immunotherapy,and further development of related targeted drugs is needed.
