摘 要:目的 探讨EB病毒(EBV)感染对结直肠癌(CRC)细胞增殖活性及T细胞平衡的影响.方法 选取2019年4月-2022年3月于福建医科大学附属龙岩市第一医院就诊的CRC患者100例为研究对象,根据EBV感染情况分为EBV感染组、无EBV感染组;比较两组患者细胞增殖迁移相关mRNA转录水平及蛋白相对表达水平;检测两组血清T淋巴细胞活化水平;将EBV与人CRC细胞株共培养后检测细胞增殖迁移活性及相关mRNA转录水平、蛋白相对表达水平,并进行分析比较.结果 与无EBV感染组比较,EBV感染组的Ki-67、P21、基质金属蛋白酶2(MMP-2)、MMP-9的mRNA转录水平与蛋白相对表达水平低(P<0.05);Treg激活水平低,Th17激活水平高,Treg/Th17低(P<0.05);EB共培养CRC细胞吸光度、迁移细胞数、迁移率、侵袭细胞数低于单独培养,且共培养细胞的Ki-67、P21、MMP-2、MMP-9的mRNA转录水平与蛋白相对表达水平低于单独培养(P<0.05).结论 EBV影响CRC细胞增殖迁移活性,可能通过影响Treg/Th17平衡介导炎性因子的释放,进而抑制CRC细胞增殖迁移.OBJECTIVE To investigate the effects of EBV infection on cell proliferation and T cell homeostasis in patients with CRC.METHODS A total of 1oo CRC patients treated in the First Hospital of Longyan City from Apr.2019 to Mar.2022 were selected as the study subjects and were divided into EBV infection group and no EBV infection group according to whether the subjects were infected EBV infection.The mRNA transcript levels and relative protein expression levels related to cell proliferation and migration were compared between the two groups.The activation level of serum T lymphocytes was detected in both groups.After co-culture of EBV with human CRC cell lines,cell proliferation and migration activity,related mRNA transcription levels,and relative protein expression levels were measured and then analyzed for comparison.RESULTS Compared with CRC patients in the no EBV infection group,the mRNA and relative protein expression levels of Ki-67,P21,MMP-2 and MMP-9 in the EBV infection group were lower(P<0.05).The activation level of Treg was lower,the activation level of Th17 was higher,and the Treg/Th17 ratio was lower(P<0.05).The absorbance,the number of migrating cells,the migration rate and the number of invading cells of EB co-culture CRC cells were lower than those of individual cultures(P<0.05),and the mRNA transcript levels and relative protein expression levels of Ki-67,P21,MMP-2 and MMP-9 of the EB co-culture CRC cells were lower than those of the individual cultures(P<0.05).CONCLUSION EBV affected the proliferation and migration activity of CRC cells,possibly by affecting the Treg/Th17 balance that mediated the release of inflammatory factors,and which in turn inhibited the proliferation and migration of CRC cells.
