摘 要:结直肠癌在我国的发病率逐年上升,但其早期诊断率较低,肿瘤异质性是结直肠癌治疗面临困难的重要原因。近年来单细胞测序(SCS)技术受到临床和科研人员高度关注,其可使研究人员能够从单细胞角度分析肿瘤细胞的异质性,并不断加深对肿瘤克隆进化机制的理解。SCS技术对于明确结直肠肿瘤亚型的发生发展、描述肿瘤微环境、研究耐药性形成机制、发现新的治疗靶点以及评估疗效与预后具有重要的生物学意义和临床价值。未来肿瘤时间与空间转变图谱的绘制实施,有助于加深对肿瘤生物学的理解,更好地精确治疗结直肠癌患者和筛选高危人群。The morbidity of colorectal cancer in China is increasing year by year,but the rate of early diagnosis is low.Tumor heterogeneity is an important reason for the difficulty of colorectal cancer treatment.In recent years,single-cell sequencing(SCS)technology has been highly concerned by clinical and scientific researchers,which enables the researchers to analyze the heterogeneity of tumor cells at the single-cell leveland continuously deepen their understanding of the mechanism of tumor cloning and evolution.SCS technology is of great biological significance and clinical value for clarifying the occurrence and development of colorectal tumor subtypes,describing the tumor microenvironment,studying the mechanism of drug resistance formation,finding new therapeutic targets and evaluating the curative effect and prognosis.The mapping of tumor transitions across space and time in the future will help deepen the understanding of tumor biology,treat the colorectal cancer patients more precisely and screen the high-risk populations.
