摘  要:为提高球孢白僵菌液体发酵产对羟基苯甲酸的能力,该研究确定了液体发酵过程中最适苯甲酸添加量为5 g/L,最优碳、氮源组合为玉米淀粉20 g/L和蛋白胨10 g/L,发酵温度为28℃,初始pH值为7.5,转速为200 r/min,接种量为17.5%。按照此条件进行验证实验,对羟基苯甲酸的产量可达3.85 g/L,产率为76.69%,与理论数据(产量为3.80 g/L,产率为75.72%)较吻合,说明此次优化方案可行。相较于优化前对羟基苯甲酸产量提升了12倍。该研究为对羟基苯甲酸的微生物合成提供了技术参考。In order to improve the ability of Beauveria bassiana to produce p-hydroxybenzoic acid by liquid fermentation,in this study,it is determined that the optimum addition amount of benzoic acid in the liquid fermentation process is 5 g/L,the optimum combination of carbon and nitrogen sources is 20 g/L corn starch and 10 g/L peptone,the fermentation temperature is 28℃,the initial pH value is 7.5,the rotation speed is 200 r/min,and the inoculation amount is 17.5%.According to these conditions,it is verified that the yield of p-hydroxybenzoic acid can reach 3.85 g/L,and the productivity is 76.69%,which are in good agreement with the theoretical data(yield of 3.80 g/L,productivity of 75.72%),indicating that the optimization scheme is feasible.Compared with that before optimization,the yield of p-hydroxybenzoic acid increases by 12 times.This study has provided technical references for the microbial synthesis of p-hydroxybenzoic acid.
