摘  要:果胶甲酯酶(pectin methylesterase, PME)是调节果胶甲酯化程度的酶,通过作用于细胞壁在植物的各个生长阶段和生理过程中发挥重要作用。目前在模式植物拟南芥中对PME研究较多,而在木本植物中研究较少。光皮桦(Betula luminifera)是中国重要用材树种,研究光皮桦PME基因家族可助于了解光皮桦木材形成机制及逆境响应机制。为研究光皮桦PME基因家族的功能,利用生物信息学方法对光皮桦基因组中PME基因家族成员进行鉴定,并对其基因组信息、蛋白生理生化特征、系统进化树、保守基序和基因表达等方面进行了研究。结果表明,在光皮桦中共鉴定88个PME蛋白序列,包括37个Type-1 PME和51个Type-2PME;预测光皮桦PME蛋白大部分为稳定的碱性亲水蛋白;光皮桦PME基因家族与毛果杨和拟南芥在聚类结果一致;光皮桦PME序列中有10个比较保守的基序,其中motif 10是Type-1 PME特有的;大多数PME基因在各组织中都有表达,尤其在雄花序中突出表达,不同Group表达情况有一定的规律性。本研究为进一步研究PME基因家族在光皮桦中的功能奠定了基础。PME(Pectin methylesterase)is an enzyme that regulates the degree of methyl esterification of pectin.It plays an important role in the various growth stages and physiological processes of plants by acting on the cell wall.At present,there are more researches on PME in the model plant Arabidopsis thaliana,but less research on woody plants.As an important timber species in China,Betula luminifera has certain research significance to study the PME gene family of B.luminifera.In order to study the function of the B.luminifera PME gene family,bioinformatics methods were used to identify the members of the PME gene family in the B.luminifera genome,and their genome information,protein physiological and biochemical characteristics,phylogenetic tree,conservative motifs and gene expression has been carried out in this research.The results showed that a total of 88 PME protein sequences were identified in B.luminifera,including 37 Type-1 PME and 51 Type-2 PME;It is predicted that most of the PME proteins of B.luminifera are stable basic hydrophilic proteins;The B.luminifera PME gene family in B.luminifera has a certain degree of conservation in protein sequence with Populus pilosa and Arabidopsis thaliana;there are 10 relatively conservative motifs in the PME sequence of B.luminifera,among which motif 10 is unique to Type-1 PME;Most PME genes are expressed in various tissues,especially prominently expressed in male inflorescence.The expression of different groups has certain regularity.This article lays the foundation for further research on the function of PME gene family in B.luminifera.
